Agilent Technologies: 25-Hydroxyvitamin D Analysis Using the Ultivo LC/TQ

The analysis of vitamin D metabolites is of interest in clinical research labs due to its role in several metabolic processes and it’s essential role in normal functions of biological systems. This fat-soluble vitamin is routinely analyzed in those labs by monitoring the metabolites 25-hydroxyvitamin D2 and D3 (25-HVD2 and 25-HVD3), but while demand for this analysis may have increased, lab space often has not.

We will present a highly sensitive and specific LC/MS analytical method developed for the Agilent Ultivo triple quadrupole (TQ) paired with a 1290 Infinity II UHPLC stack, the smallest footprint LC/TQ system available on the market. A simple gradient is used to separate the metabolites, with all monitored analytes eluting in under 5 minutes. Excellent linearity over three orders of magnitude is demonstrated and good peak shape and reproducibility is demonstrated at all levels providing a robust and reliable quantitation.

Andre graduated from University of Illinois in Urbana-Champaign with a master’s degree in Food Science. Working for over 30 years in the instrumentation industry as an applications chemist, Andre has specialized in the development and implementation of LC-MS applications for various industries including clinical research. He has expert knowledge of chromatography (from nano- to preparative-scale) and mass spectrometry with extensive practical knowledge in areas of small and large molecules, quantitation, proteomics, metabolites identification, forensics, clinical, toxicology, food and environmental analysis.
