There are many things learned over a career in practicing liquid chromatography in the analytical laboratory. This webinar is intended to be a sharing of suggested and recommended practices for various aspects of carrying out experiments and method development for HPLC and UHPLC. It will include a review of some tools and practices that can make column selection, usage, flushing, cleaning, and storage easier and more successful. Other types of measurements such as instrument dead volume, extracolumn volume, and extracolumn dispersion will also be described.

We will also discuss some method development strategies that may be beneficial in helping you to be more productive, including some examples that demonstrate the successful use of DryLab software for method development and optimization. In addition, some specific examples will be discussed that describe practical learnings from several situations that occurred in a 37-year (and counting) career in the chemical industry.

There will be an opportunity for some questions and answers, and any questions, comments and suggestions from attendees that are not answered live will be answered by email after the webinar.

  • Do’s and Don’ts for pH meter calibration and measurement, including the use of control solutions for quality assurance.
  • How do you know which stationary phase is going to work for your current or next method development?
  • How to select similar and dissimilar (orthogonal) columns for method development and for drop-in as alternative (equivalent) for a column for an existing method?
  • How to estimate expected backpressure for a column geometry before you purchase it
  • How to measure extracolumn dispersion and assess its impact when using different column geometries
  • How to use mobile phase pH for method development, and how to use mid- and high pH for compatible columns
