Agilent Technologies: Avoid System Contamination with Ion-Pair-Free Analysis of Oligonucleotides by LC and RapidFire HILIC MS

LC/MS methods for the analysis of oligonucleotides have traditionally been based on ion-pairing reverse-phase (IP-RP) chromatography because these approaches generally deliver good separation and MS response in negative mode. However, considering that many ion-pairing reagents can present a memory effect which negatively impacts the performance of the system in positive mode, IP-RP methods can be burdensome for mixed-use systems and push many laboratories to seek chromatographic alternatives.

In this webinar, we present high-quality MS analysis of a wide variety of oligonucleotide samples without the use of ion-pairing reagents, on both LC and RapidFire 6545XT AdvanceBio QTOF configurations. These methods leverage Agilent HILIC-Z resins and MS-friendly, ammonium acetate-based, mobile phases, which allow for subsequent positive mode use of the system with little to no flushing or hardware changes.
