Using an in-house UNIFI library with the ACQUITY RDa Detector to Determine Absolute Configurations of Amino Acids in Natural Products

The absolute configuration of the α-carbon (C-2) in amino acids is critical in the characterization of peptides. We developed an optimized analysis of Marfey’s method, a popular absolute configuration technique that compares retention times of derivatized hydrolysates to commercial ʟ,ᴅ standards. The improved analyses utilizes an in-house Waters UNIFI library in tandem with the ACQUITY RDa Detector, a high-resolution time-of-flight mass analyzer. The UNIFI library of FDVA-derivatized amino acids expedites the Marfey’s application by simultaneously acquiring data and processing samples for high-throughput determination of the α-carbon absolute configuration. This easily tailored workflow allows analysis of peptidic small molecules more efficiently.

Conformation-Specific Native Top-down Mass Spectrometry to Study Dynamic Protein Complexes and Conformations

Native top-down mass spectrometry is a powerful tool for analysis of proteins and their complexes, characterizing backbone organization, interaction interfaces, solvent accessibility, and post-translational modifications. To date, such experiments have sampled across the entire conformational landscape, therefore the results reflect an average across multiple protein structures which cannot be delineated. Here, we combine electron capture dissociation with cyclic ion mobility-mass spectrometry to perform native top-down MS in a conformation-specific manner. Having successfully developed this method, it is now being applied to study protein conformation in cases of diseases known to be caused by misfolding or disorder of challenging proteins.

Desorption Electrospray Ionization - Mass Spectrometry Imaging and Cyclic Ion Mobility for the Study of Lipids in the Injured Brain

Desorption Electrospray Ionization (DESI) has emerged as a powerful ionization technique for lipid imaging. However, the complexity of lipid species, structures, and ionization make lipids difficult to study without any additional dimension of separation. An ion mobility separation effected by a cyclic ion mobility (cIM) device can declutter mass spectra from complex biological samples, separate isobaric and isomeric species and facilitate the identification of lipids. DESI-MSI for the study of lipids coupled to cIM separation will be discussed as well as the improvement from the new source (DESI-XS) and TOF analyser (Select Series).
