Unleashing the Potential of Bravo Automation
The Agilent Bravo is a flexible liquid handling platform that automates sample preparation for screening applications such as compound management, cell-based assays, and biochemical assays. With this robotic liquid handling system, time is freed up and consistent data are achieved across samples and users. Utilizing the powerful VWorks software and application-based collection of pre-prepared protocols the Bravo can be applied to the basic as well as the very complex workflows by different levels of end users. Combining the power of the software with different pipetting heads and on deck accessories allows the Bravo to be configured for workflows such as proteomics, metabolomics, lipidomics and genomics.

Presenter: Kenda LJ Evans, PhD (Automation Product Specialist, Agilent Technologies, Inc.)
Kenda received her Ph.D. from The University of Texas Health Science Center in San Antonio in the field of Pharmacology with a focus on the signal transduction pathway for the Serotonin 1A (5-HT1A) receptor. Kenda’s post-doctoral work occurred at the University of Houston School of Pharmacy where she studied the effects of inverse agonists and antagonists on Beta receptors in both respiratory and cardiovascular models. Kenda then joined Encysive Pharmaceuticals where she was the lead for the high throughput screening team as well as project manager for target drugs. Following this, Kenda joined PerkinElmer as a high throughput screening Field Application Scientist. For more than eleven years now, Kenda has been an Automation Product Specialist for Agilent where she is helping researchers find the right solution for their automation needs.
Decoding Automation of Metabolite and Lipid Extraction Workflows
Technology improvements in liquid chromatography/mass spectrometry have enhanced the detection and identification of metabolites and lipids from complex biological samples. As metabolomics and lipidomics measurements become increasingly valued, there is a growing need to automate sample preparation workflows. Specifically, Agilent automation offers intuitive workflows that provide high data reproducibility and increased throughput while reducing hands-on time. Here, we describe key learnings revealed during the automation of several workflows that extract metabolites and/or lipids from plasma and mammalian cell samples.

Genevieve Van de Bittner joined Agilent Research Laboratories in 2016 with a goal of improving measurement technologies. Toward this goal, Genevieve has developed automated sample preparation methods for LC/MS metabolomic and lipidomic analyses and explored the combination of Seahorse and Mass Spectrometry measurements for developing a more comprehensive understanding of cellular biology. These efforts have been informed by Genevieve’s training in chemical biology, cell analysis, and clinical research at UC Berkeley and Harvard Medical School.
Sample Preparation for Phosphoproteomics, Immunopeptidomics, Protein-Protein interaction studies and more on a Single Automated Platform
The AssayMAP Bravo is a flexible yet easy-to-use automated microchromatography platform that leverages packed resin bed cartridges, positive-pressure displacement, precision flow control, and preoptimized applications to perform a wide variety of protein sample preparation workflows required for proteomic studies and biotherapeutic development. The automation of protein sample preparation workflows minimizes variability, manual labor, and human error. In addition, the ability to process up to 96 samples in parallel enables proteomic analysis at a scale that was previously impractical. This presentation will demonstrate how this platform enables multiple proteomic approaches such as phosphoproteomics, immunopeptidomics, and protein-protein interaction mapping.

Steve earned his Ph.D. at the University of California, San Francisco and did his post-doctoral work at Stanford University. Following his post-doctoral work, Steve worked at multiple biotechnology companies developing, running, and manufacturing high throughput drug discovery assays. For the last 12 years, Steve has been developing automated protein sample preparation workflows for protein quantitation and characterization on the AssayMAP Bravo platform.
