The development of chromatographic assays for analytes in biological fluids can be a daunting challenge due to the complexity of sample matrices and analyte chemistries. However, method development challenges can be broadly overcome with better understanding of the parameters involved. When dealing with sample preparation it is important to not only provide consideration to the analytes of interest but also to carefully investigate the chosen matrix.

In this seminar, we will discuss some key considerations involved in developing successful sample preparation methods for extracting analytes from various clinical matrices. Emphasis will be placed on experimental design, analyte and matrix properties and choice of extraction approach. We will touch on common sample prep techniques such as Supported Liquid Extraction and Solid-Phase Extraction illustrating fundamentals for use, chemical interactions, and keys for success.

Presenter: Lee Williams, PhD

Lee gained his Ph.D. in mass spectrometry under the tutorship of Prof. David E. Games from the University of Wales, Swansea in 2000. He went on to complete a Post-Doc in the Biochemical Toxicology department at the National Center for Toxicological Research, FDA labs in Arkansas, USA. In 2004 he returned to Wales to join Biotage® (Formerly Argonaut) to advance the LC/MS capability within the sample preparation arena. Over the 16 years, he has occupied a variety of roles from senior scientist, group leader, lab manager and most recently Director of R&D for Analytical Consumables and Applications.
