Thermo Scientific: A new approach to perform ultra-sensitive, accurate, and absolute quantification of key membrane receptors and antigens in tissue and on cell surfaces

Developing targeted LC-MS/MS work to quantify key membrane receptors and antigens to evaluate drug targets and the effect of biotherapeutics is extremely challenging.

The expression levels of these proteins are often exceptionally low, the properties of these membrane proteins can limit the effectiveness of extraction and recovery, and the noises from biological matrices are typically high. Traditional methods often suffer from insufficient sensitivity and quantitative accuracy.

The focus of this webinar is to introduce an alternative method designed to implement more generic sample preparation steps in combination of strategic trap-elute chromatography, FAIMS selectivity, and sensitive LC-MS/MS acquisition utilizing selected reaction monitoring (SRM) on a premier triple quadrupole mass spectrometer. Prof. Qu and his team will demonstrate confident quantitation of various membrane receptors and antigens that are important to the biotherapeutics. The presented workflow will also demonstrate advantages to streamlining method development to increase productivity.

Presenter: Prof. Jun Qu (Assistant Professor, State University New York (SUNY) Buffalo)

Jun Qu is a professor in the Department of Pharmaceutical Sciences of SUNY-Buffalo, and the director of Proteomics and Pharmaceutical Analysis Group in NY Center of Excellence in Life Sciences. His research is focused on the study of Clinical/Pharmaceutical Proteomics and Pharmaceutical Analysis by developing novel LC/MS-based strategies to markedly enhance the sensitivity, specificity, throughput and accuracy/precision for protein analysis in highly complex biological matrices. His research programs include i) highly sensitive and accurate characterization of biotherapeutics, targets and markers in pharmaceutical and clinical systems; ii) precise and large-scale global quantification of pathological proteomes using a novel MS1-based proteomic pipeline (IonStar); iii) sensitive identification, localization and quantification of post-translational modifications in tissue proteomes; iv) targeted investigation of marker proteins that are of high interests for clinical and pharmaceutical investigations. Qu lab is also one of the world leaders in LC-MS-based characterization of biotherapeutics in pre-clinical models. He has around 130 peer reviewed publications and >70 grants funded as the principal investigator or co-investigator.
