Size-exclusion chromatography coupled with multi-angle light scattering (SEC-MALS) has emerged as a powerful tool for comprehensive protein characterization over the past decade. Unlike mass spectrometry, SEC-MALS analyzes proteins in their native solution state, offering greater ease of use and affordability. This versatility has made SEC-MALS a preferred platform for routine analysis of multiple quality attributes in a single measurement throughout the protein product lifecycle.

During the early discovery stage, SEC-MALS provides valuable insights into protein oligomeric state, conjugate composition, aggregation content, shape, and extinction coefficient. As the protein therapeutic program advances to process and analytical development, SEC-MALS facilitates rapid assessments of aggregate size and abundance. In QAQC of complex protein therapeutics, such as heavily glycosylated proteins and PEGylated proteins, SEC-MALS often serves as a release assay for both molecular weight distribution and composition. For non-conjugated proteins, SEC-MALS frequently complements the release panel to evaluate lot and process comparability. This webinar will explore the diverse applications of SEC-MALS and related light scattering techniques in analyzing proteins for their various quality attributes.
