Native size-exclusion chromatography coupled to mass-spectrometry (nSEC-MS) is a suitable tool for assessment of protein quality, i.e. purity and molecular weight.

SEC alone can be considered as straightforward and easy, given that it is typically performed with isocratic elution at constant flow and ambient temperature, where the separation is ideally only dependent on size (hydrodynamic radius). However, obtaining good chromatography places a high demand on using optimal mobile phases and bioinert columns that limits any secondary interactions without altering the sample purity profile. Coupling SEC to mass-spectrometry makes this even more challenging, since volatile mobile phases are required. This webinar shows 1) tips to setting up nSEC-MS on BioAccord LC-MS, 2) data generated and lessons learned from case studies, and 3) method optimization and options do consider depending on your analytical needs.
