RT-MALS: Real-Time PAT for biologics, gene vectors and vaccines in downstream processing

While much effort has been invested in process analytics for upstream bioprocess, downstream operations still suffer from a lack of technologies suitable for inline/online monitoring of product attributes with rapid response times. Such a technology is real-time multi-angle light scattering (RT-MALS), which can be integrated inline with chromatographic purification or online with UF/DF and homogenization equipment.
RT-MALS determines biophysical attributes such as molecular weight and size of proteins, nucleic acids and polysaccharides, as well as size and particle concentration of lipid nanoparticles or viral vectors. Molecular weight and size discriminate product from impurities such as residual DNA or aggregates. For small vectors such as AAV, RT-MALS also determines empty and full titers and the full:total capsid ratio.
