The Problem

Characterizing ADCs can feel like reinventing the wheel when methods aren’t flexible enough to keep up with high-throughput analysis of different antibody-drug combinations or reaction conditions. ADC characterization usually involves a combination of several HPLC methods, but HPLC often takes too long, requires too much sample, or needs too much optimization to work with every candidate.

The Solution

Stunner makes high-throughput reads on 2 μL of sample to gather three key datapoints on up to 96 ADCs at-a-time. Keep an eye on antibody concentration, check the drug-antibody ratio (DAR), and watch out for aggregation – all in only two minutes per sample.

The Proof

In this seminar, we’ll show how Stunner characterized 6 model ADCs created from 1 mAb and 3 payload/linker combinations reacted at 2 molar ratios. We’ll explain how Stunner’s Unmix algorithms break down UV/Vis absorbance spectra into their component parts to quantify total protein and drug payload. We’ll also show how flexible that analysis is when it is taught new user-created reference spectra.

His expertise includes biophysical characterization of viral vectors, lipid nanoparticles, proteins, and nucleic acids. Prior to Unchained Labs, his research experience covered the fields of sustained antibody delivery and nanotechnology. He earned his Ph.D. in Bioengineering from the joint UC Berkeley – UCSF Bioengineering Graduate Program.
