Agilent Technologies: LC Method Transfer: Hidden Challenges & Absolute Solution

A real challenge in chromatography labs today is that the method transfer between instruments is rarely as simple as it sounds because of the many variables involved. Older LC systems like Agilent 1100/1200/1260, or any other known make legacy instrument, which often differ significantly in hardware, dwell volume, gradient delay, and pump technology compared to modern systems like Agilent 1260 Infinity III Prime or 1290 Infinity III LC.

Why Method Transfer Is Complex:

  1. Instrument Variables: Gradient delay volume, pump mixing type (low-pressure vs high-pressure), detector cell volume, tubing dimensions, and even software algorithms can impact retention times and peak shapes.
  2. Legacy Methods: Many older methods were optimized for specific hardware quirks, making direct transfer risky without adjustments.

Sambasivarao holds a Master of Pharmacy degree from SRM University, Chennai, and has 15 years of industry experience. He spent a decade working in New Product Development and Analytical Research & Development at leading pharmaceutical companies such as Syngene and MicroLabs.

Currently, he serves as an Application Engineer at Agilent Technologies, specializing in Chromatography, Dissolution, and Laboratory Informatics, with five years of experience in this role. His areas of expertise include Chromatography, Quality by Design (QbD), and Purification techniques.
