Agilent Technologies: Scale Up in Minutes: A Practical Example of Preparative HPLC Scale Up from an Analytical Method

Preparative liquid chromatography begins with an analytical separation. First, we need to confirm the presence of the target compound in the sample. Then, we must assess whether the estimated amount of the target compound we can recover from the sample justifies the subsequent purification effort. Preparative chromatography is not based on guesswork but is founded solidly on a set of well-documented rules to be followed for optimum results when scaled from an analytical method.

Scouting for appropriate starting conditions, optimizing for speed, yield, and purity are fundamental considerations. The desired sample throughput determines priorities: high yields for a few different samples justify optimization of yield, whereas dealing with large numbers of different samples at the milligram scale demands proper automation.

The examples will utilize practical considerations such as focused method development, loading studies and understanding the principles of scale up. The Preparative HPLC Calculator is one of the tools available that was developed to improve the ease with which you can scale up your analytical methods to meet your separation goals. We will utilize a Preparative Calculator tool to quickly determine the conditions for our preparative separation with predictable results that will meet the goals of our projects.

Presenter: Lori Sandford (HPLC Applications Scientist, Agilent Technologies, Inc.)

Lori Sandford is an HPLC Applications Scientist with Agilent Technologies, Inc. in Wood Dale, IL. Lori has been working at Agilent Technologies supporting HPLC instrumentation since 2010, including 2 years with Varian working as an HPLC Specialist.

Prior to Agilent, she was the Global Technology Manager for Prep and Process Chromatography at W. R. Grace/Alltech. Her career started at G.D. Searle/Pfizer in 1998 as a chromatography specialist in the Prep and Process Chromatography group supporting both Medicinal and Process Separations. Lori attended Florida State University where she received her degree in Chemistry. When not tinkering in the lab, Lori is enjoys time at home with her 2 teenage boys shuttling from hockey rink to Lacrosse field, enjoying concerts or planning her next scuba adventure.

Presenter: Lorena Lopez Pena (LC Application Scientist, Agilent Technologies, Inc.)

Lorena has a degree in Chemical Engineering, and she is currently based out of the Agilent facilities in Lexington, Massachusetts. She has been with Agilent for almost 7 years tightly involved within Liquid Chromatography and Dissolution instrumentation.

She has a strong background in the pharmaceutical industry where she started her career as a lab scientist, followed by scientific research and development, and has been focusing on the Biopharma field for the last couple of years within her current role as an LC Application Scientist.
