Rapid Detection of Undeclared Active Ingredients in Online Health Supplements Using DART (Direct Analysis in Real Time) Open Ambient Ionization Source Coupled to ACQUITY QDa Mass Detector

• Royal Honey: A tert-butyl ether “solvent swap” removed polar sugars, revealing tadalafil (m/z 390) and characteristic fragments (m/z 268, 135) matching the standard.
• Nuvitra King Capsule: Direct OpenSpot analysis detected sibutramine and fluoxetine. Elevating DART temperature enhanced ionization of fluoxetine and produced fragment ions at m/z 125 and 195 for sibutramine.
• Ortiga Joint Remedy Tablets: Contrary to FDA suspicions of diclofenac, analysis showed acetaminophen (m/z 152) with fragment m/z 110, consistent with the acetaminophen standard.

These findings illustrate the DART QDa system’s capability to screen multiple dosage forms and complex matrices with high specificity.
