We observed that the transcription start site can vary on the basis of the compatibility of the specific template and dinucleoside polyphosphate. Cryo-electron microscopy structures of transcription initiation complexes with dinucleoside polyphosphates revealed that both nucleobase moieties can pair with the DNA template. The first encoded nucleotide pairs in a canonical Watson–Crick manner, whereas the second nucleobase pairs noncanonically in a reverse Watson–Crick manner. Some scientists have even experienced total analyte loss with samples stored in vials for an extended time. The vial chemistry, specifically the&nbsp;glass surface, is the major contributor to this problem.</p><p>Polar analytes&nbsp;may adsorb to the glass vial surface by polar-polar interaction and/or ion exchange mechanisms, thereby reducing the overall amount of sample in solution. TruView Certified LCMS Vials&nbsp;reduce this problem dramatically due to a stringent manufacturing process.
