The research group of associate professor Bob Pirok at the University of Amsterdam’s Van ’t Hoff Institute for Molecular Sciences (HIMS) is partnering with Agilent Technologies to develop advanced analytical methodologies for polymer characterisation. An example is the STREAMLINED consortium led by Pirok and his HIMS colleague Dr Alina Astefanei, developing analytical tools to investigate nanoplastics. Coupling mass spectrometry to liquid chromatography

This session will outline the factors which impact LC/MS methods, such as solvents and additives.

Light scattering techniques deliver multi-attribute quantification (MAQ) of mRNA and lipid nanoparticle-formulated mRNA and can play a key role in product and process development as well as quality control of mRNA-LNP therapeutics. Specifically, multi-angle light scattering (MALS) coupled to separation by field-flow fractionation (FFF) or size-exclusion chromatography (SEC) provides high-resolution quantification of several mRNA and LNP attributes including size distribution, physical stability, LNP number concentration, morphology, and mRNA and lipid concentration.
The method can also be applied to evaluate how these attributes are affected by preparation, formulation, and storage conditions. In addition to high-resolution MAQ by SEC/FFF-MALS, dynamic light scattering (DLS) and electrophoretic light scattering (ELS) can be used for rapid, automatable batch measurements of particle size and concentration, polydispersity, and zeta potential, enabling rapid screening of the impact of different ions, ionic strength, pH, temperature and other parameters on the biophysical properties and stability of mRNA and LNP-mRNA.

His postdoctoral research in protein folding and misfolding biophysics focused on understanding and preventing misfolding in bulk solution and on artificial surfaces used in medicine and biotechnology, and he has also developed protein- and aptamer-based electrochemical biosensors for continuous real-time measurements in vitro and in vivo. Martin received his M.Sc. in Chemistry and Ph.D. in Biochemistry from Stockholm University, Sweden.

Sample Preparation: Theory and Overview

The first episode will introduce you into goals and available techniques of Solid Phase Extraction (SPE).

Chemical recycling holds the key to keeping plastics in the loop. Plastic waste is chemically converted into substances that can be reused as raw materials for plastic production. Pyrolysis oil is an intermediate product during this process. Among its main contaminants are nitrogen, sulfur, and halogens, which can cause fouling, corrosion, and catalyst poisoning during further processing. Therefore, quality control is essential to optimize treatment and prevent emissions, extended maintenance cycles, and low-quality hydrocarbon products.
The complexity of chemical recycling is influenced by the raw material entering the process. Plastic waste can have a high diversity in chemical composition and physical properties, resulting in a high variability of the pyrolysis oil. Thus, one of the biggest challenges in analyzing pyrolysis oil is handling the different product grades – from low boilers to heavy cuts. Furthermore, samples can be inhomogeneous, highly viscous, and vigorously reacting, which makes it difficult to ensure correct analysis results.
The analysis by pyrolytic and pyrohydrolytic combustion has proven as best suited for the quick and combined determination of all relevant non-metal elements over a wide concentration range. The presented instruments ensure safe, soot-free, and quantitative combustion with short analysis times. Furthermore, the instruments can not only be utilized for the analysis of pyrolysis oil, but also for the analysis of the plastics entering the chemical recycling process.
