Meeting EU Drinking Water Standards: The Critical Role of Sample Prep in Trace Contaminant Analysis

Although modern agriculture needs to be based on accurate real-time data on soil, plant, and environmental conditions, affordable, sufficiently sensitive, and robust monitoring devices for this purpose are currently missing. According to scientists, low-cost sensors based on new smart materials and printing technologies offer a suitable solution. The results of multidisciplinary research thus far and proposals for the development of these devices have been presented by researchers from Czechia, Great Britain, Spain, and the USA in a review article in the prestigious journal <a target="_blank" rel="noopener noreferrer" href="https://www.nature.com/articles/s41467-026-68778-6#Abs1">Nature Communications</a>.“Sensor technologies for monitoring plant health are still in their infancy. Only simple sensors capable of monitoring basic parameters such as pH, soil moisture, or temperature are commonly available on the market. However, if farmers are to face current challenges, including climate change, and ensure sufficient food for humanity, they need much more effective tools that will enable them to monitor, for example, various pathogens, including bacteria and fungi. Materials research and the use of graphene-based smart materials were led by scientists from CATRIN at Palacký University and VŠB-Technical University of Ostrava. Monoclonal antibodies (mAbs) and their derivative products are a very important class of biopharmaceutical molecules. mAbs are tetrameric glycoproteins having a molecular mass of approximately 150 kDa, composed of two heavy chains and two light chains, inter-linked by several disulfide bonds, and having at least one conserved N-glycosylation site located in the Fc domain. mAbs were introduced for the treatment of various diseases in the late 1980 and they still represent the most rapidly growing category of therapeutic molecules today. There are more than one hundred mAbs approved by the US Food and Drug Administration (FDA). mAb are particularly interesting because of their good therapeutic efficiency, favorable pharmacokinetic (PK) and pharmacodynamics (PD), and relatively low side-effects.

Welcome to the 2022 Detection and Quantification of PFAS in Air, Water, Soil, and Food Packaging e-learning series.
In the third event of our three-part eSeminar series, a leading expert in PFAS analysis will discuss a variety of topics around PFAS in food packaging materials, including method development and sample preparation.
A non-intentionally added substance or NIAS, in packaging is an issue of growing concern. First mentioned in EU Regulation 10/2011, predictable and unpredictable NIAS, in the form of degradation products and impurities, were found not only in plastic but also in paper and in newly developed materials, such as recycled and biobased packaging.
In the case of recycled materials, there is a new type of NIAS, which comes from recycling cycles: an accumulation of different mixture of chemicals might occur when materials are recycled many times. The prediction, identification and control of NIAS in recycled materials is therefore challenging because of the difficulty tracing their origin.
Due to the complex identification of the contamination pathway, chemicals like PFAS can be considered to be both intentionally and non-intentionally added. Some PFAS molecules are included for use and find market applications in food contact materials due to their grease- and water-proof properties. In this case they are intentionally added. This is frequent in paper packaging; and when this also has a recycled content, PFAS can be present as NIAS, originating from the intentionally added PFAS in the recycled paper fibres, or from inks, for example. 
Environmental contamination should also not be ignored, where PFAS can be introduced in paper packaging from water used during the recycling or manufacturing process or from the atmosphere. 
Distinguishing between these sources is important: new instrumental technologies and approaches are needed, not only for analytical efficiency and for performance, but also to open up the identification and quantification of what is predictable and what is not.

PFAS: A package deal

We will discuss a variety of topics around PFAS in food packaging materials, including method development and sample preparation.

In 2018 Valsartan was found to contain the N-Nitrosamine, NDMA. This discovery set in motion an issue that, still to this day, has sent shock waves through the Pharmaceutical Industry and continues to threaten the future viability of many critical medicines.
This presentation will seek to examine the chain of events that have led us to the present day and asks the question: Is the issue we now faced the same issue as that first identified in the Valsartan incident and are the same approaches valid?
The evolution of the issue from a drug substance contamination issue to one related to N-Nitrosamines formed from the active itself. How the two issues are distinct and how by treating them as equivalent threatens the very existence of many critical medicines and the choices industry and regulators now face.

Metabolomics with MALDI Imaging Mass Spectrometry

Lipid nanoparticles (LNPs) have been proven to be highly effective delivery vehicles for nucleic acid-based therapeutics. As multi-component delivery vehicles, LNPs are comprised of four lipid components, each with a specific function and requiring a specific ratio to effectively and safely deliver the nucleic acid payload (mRNA, siRNA guide RNA, etc.). Recent studies have shown that some lipid impurities associated with ionic lipids in particular can result in lipid adduct formation on mRNA that negatively impacts the safety and efficacy of the final drug product, highlighting the need for robust LNP composition and impurities analysis.
LC-based methods are frequently used as the principal platform in the analysis of LNPs. MS-based methods tend to be deployed in upstream activity for characterization and identification purposes, whereas optical-based methods using detectors such as ELSD are deployed in late-stage development or manufacturing environments as part of monitoring or screening assays.
In this webinar, LC-ELSD and LC-ELSD-MS workflows that can be run under Empower CDS for easy deployment in regulated and non-regulated labs will be reviewed along with LNP composition and impurities analysis data. Guidance will be provided as well on ELSD parameters that can be adjusted for optimum results.
