Negative Mode Detection of Pigments in Cosmetics using the MALDI-8030 Dual Polarity Benchtop MALDI-TOF Mass Spectrometer
Applications | 2022 | ShimadzuInstrumentation
Cosmetic pigments play a critical role in product appeal and safety in a market exceeding USD 863 billion. Reliable analytical methods ensure regulatory compliance, ingredient transparency and support consumer choices related to vegan, organic and halal standards.
This study evaluates a rapid workflow using a dual-polarity benchtop MALDI-8030 linear TOF mass spectrometer in negative ion mode for qualitative detection of pigments in lipstick formulations. Four commercial samples—non-vegan/non-organic, organic (non-vegan), vegan/halal (non-organic) and a second non-vegan/non-organic shade—were selected to represent key market segments.
Samples of each lipstick were extracted by adding a 1:1 water/methanol mixture, sonicating for 30 minutes and centrifuging to recover pigment-rich supernatant. Extracts were mixed with 9-aminoacridine matrix and spotted for MALDI analysis in negative ion mode. MS/MS was performed on a higher-resolution instrument to confirm standard identities but is not required for routine screening.
Non-vegan/non-organic lipsticks showed expected peaks for Red 7 (CI 15850), Yellow 6 Lake (CI 15985) and, in one shade, Yellow 5 Lake (CI 19140). An organic non-vegan lipstick yielded a clear Carmine (CI 75470) signal, while the vegan/halal sample contained CI 12085 and CI 15850:1 without Carmine. Isotopic patterns of brominated pigments and MS/MS spectra provided unambiguous identification.
Advancements may include quantitative MALDI imaging of pigment distribution, expansion to other personal care matrices and integration with automated sample handling. Emerging matrices and ambient ionization could further reduce prep time and broaden applicability to in-field testing.
The MALDI-8030 benchtop system combined with a straightforward extraction protocol provides a reliable qualitative screen for a range of cosmetic pigments. This approach supports regulatory requirements and market demands for ethical ingredient sourcing.

Summary
Importance of the Topic
Cosmetic pigments play a critical role in product appeal and safety in a market exceeding USD 863 billion. Reliable analytical methods ensure regulatory compliance, ingredient transparency and support consumer choices related to vegan, organic and halal standards.
Objectives and Study Overview
This study evaluates a rapid workflow using a dual-polarity benchtop MALDI-8030 linear TOF mass spectrometer in negative ion mode for qualitative detection of pigments in lipstick formulations. Four commercial samples—non-vegan/non-organic, organic (non-vegan), vegan/halal (non-organic) and a second non-vegan/non-organic shade—were selected to represent key market segments.
Methodology
Samples of each lipstick were extracted by adding a 1:1 water/methanol mixture, sonicating for 30 minutes and centrifuging to recover pigment-rich supernatant. Extracts were mixed with 9-aminoacridine matrix and spotted for MALDI analysis in negative ion mode. MS/MS was performed on a higher-resolution instrument to confirm standard identities but is not required for routine screening.
Instrumentation Used
- MALDI-8030 dual-polarity benchtop linear MALDI-TOF mass spectrometer
- 9-Aminoacridine matrix (10 mg/mL in methanol)
- Ultrasonic bath and microcentrifuge
Main Results and Discussion
Non-vegan/non-organic lipsticks showed expected peaks for Red 7 (CI 15850), Yellow 6 Lake (CI 15985) and, in one shade, Yellow 5 Lake (CI 19140). An organic non-vegan lipstick yielded a clear Carmine (CI 75470) signal, while the vegan/halal sample contained CI 12085 and CI 15850:1 without Carmine. Isotopic patterns of brominated pigments and MS/MS spectra provided unambiguous identification.
Benefits and Practical Applications
- Simple, rapid sample preparation without extensive cleanup
- High resolution and accurate mass detection of water-insoluble pigment lakes
- Capability to verify compliance with vegan, organic and halal claims
- Affordable benchtop platform suitable for QA/QC in cosmetic laboratories
Future Trends and Opportunities
Advancements may include quantitative MALDI imaging of pigment distribution, expansion to other personal care matrices and integration with automated sample handling. Emerging matrices and ambient ionization could further reduce prep time and broaden applicability to in-field testing.
Conclusion
The MALDI-8030 benchtop system combined with a straightforward extraction protocol provides a reliable qualitative screen for a range of cosmetic pigments. This approach supports regulatory requirements and market demands for ethical ingredient sourcing.
