And while all non-volatiles give a similar uniform response, <a target="_blank" rel="noopener noreferrer" href="https://www.thermofisher.com/content/lifetech/global/en/home/industrial/chromatography/chromatography-learning-center/liquid-chromatography-information/hplc-system-components/how-hplc-detectors-work/charged-aerosol-detection/charged-aerosol-detection-cad-performance-tips.html">the response of semi-volatiles</a> depends on volatility and consequently is less predictable.So, the question then becomes is there a way to address this performance difference and make semi-volatile detection more predictable? One approach for charged semi-volatile analytes is to add a volatile ion-pairing agent of the opposite charge in the mobile phase to force non-volatile behavior.However, this method is analyte specific and may also react with impurities in your mobile phase, leading to higher noise and poorer signal-to-noise (S/N) ratio. What’s really needed is a more global approach to improve the accuracy for measuring semi-volatiles.This approach is now achievable with Temperature Coupling Mode (TCM) in the <a target="_blank" rel="noopener noreferrer" href="https://discover-pharma.com/thermo-fisher-upgrades-charged-aerosol-detection-tech-to-improve-complex-drug-analysis/">Thermo Scientific Vanquish CAD P Series</a>.<h3>Why stricter temperature control matters for measuring semi-volatiles</h3>Before we get into the specifics, you need to understand why temperature control in the CAD is critical to quantitative accuracy, especially for semi-volatiles (Figure 1).<img src="https://storage.googleapis.com/labrulez-bucket-strapi-h3hsga3/Thermo_Fisher_Scientific_How_to_Get_More_Accurate_HPLC_Quantitation_of_Semi_Volatiles_2_e33cff4cac.jpeg" alt="Thermo Fisher Scientific: How to Get More Accurate HPLC Quantitation of Semi-Volatiles: Figure 1. This fixed temperature difference between the EvapT and CDM could negatively impact the measurement of semi-volatile compounds that require a lower EvapT.Adding TCM resolves this issue, by linking the EvapT and CDM temperatures, thereby improving the quantitation of semi-volatile species.<h3>Using TCM to help improve quantification of semi-volatiles, like fatty acids</h3>To demonstrate the effectiveness of this new temperature coupling feature, we anlazyed a working solution of four individual fatty acids (FAs) and compared the CAD repsonse with TCM turned on and turned off (Figure 2).<img src="https://storage.googleapis.com/labrulez-bucket-strapi-h3hsga3/Thermo_Fisher_Scientific_How_to_Get_More_Accurate_HPLC_Quantitation_of_Semi_Volatiles_3_da6cf5006c.jpeg" alt="Thermo Fisher Scientific: How to Get More Accurate HPLC Quantitation of Semi-Volatiles: Figure 2. Separchrom PC01 ECOM industrial preparative columns correspond to the highest requirements of preparative liquid chromatography. They are designed to be used in a mode of axial compression with a moving input piston.

They are designed to be used in a mode of axial compression with a moving input piston. In the original design, they are equipped with identical pistons on input and output (biaxial compression as an option). In the ECOM Separchrom PC FE modification, they have a fixed end plate on the output instead of the piston. Pistons and plates are equipped with frits. A liquid distributing system is installed under each frit. The design guarantees perfect piston flow, high efficiency even for high injection volume, and simple accessibility of frits for cleaning. Columns can be packed by dynamic slurry method or by sedimentation or with sorbent in a dry state.SEPARCHROM PC 01 ECOM columns are made of stainless steel AISI 316L (tube inner surface is polished to reach Ra &lt; 0.3 µm) with stainless steel pistons and UHMWPE (PTFE on the request) sealing rings. The stroke of the main piston is proportional to the used hydraulic cylinder or flange bolt length. Pistons are sectional; the frit (sintered mesh disc 3 µm pore size, 5 mm thickness) with the frit ring can be removed. The output plate in the FE version is made of UHMWPE, frit is fixed with a simple frit ring. Column flanges are not welded, but screwed onto the column tube. No temperature increase during manufacturing means no change in steel properties. Flow distributor with groves is incorporated into the piston to guarantee piston flow with nearly zero pressure drop. The design guarantees simple accessibility of frits for cleaning.Columns can be packed by dynamic slurry method, or by sedimentation, or with sorbent in the dry state, both with silica and polymer sorbents.
