At the January 2025 CFSRE Seized Drugs Symposium, a Thermo Fisher Scientific–sponsored workshop showcased the potential of <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/2240">Thermo Scientific VeriSpray PaperSpray mass spectrometry </a>(PS-MS) to transform drug analysis workflows in both forensic and clinical toxicology. Stefania’s PhD research seeks to change that. Her team is developing a rapid, extraction-free method using VeriSpray PS-MS for the targeted analysis of plasma samples from patients presenting with acute drug toxicity.By using dried matrix spots on a paper substrate and coupling this with a <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/404">Thermo Scientific TSQ Altis Plus Triple Quadrupole Mass Spectrometer</a>, Stefania’s work provides an alternative screening technique to chromatography, delivering faster, actionable results to guide early clinical decision-making.<h5>Key takeaways:</h5><ul><li>Paper spray MS is an ambient ionization technique suitable for emergency toxicology due to its rapid results and minimal sample prep.</li><li>Enhancing the paper substrate and adding microextraction steps can boost sensitivity without sacrificing speed.</li></ul><h3>Substrate matters: boosting detection with paper treatment</h3>One key limitation of PS-MS is the interference that can arise from the paper substrate itself. Marwa’s research explored nine different paper treatments, ranging from acids to solvents, to improve the signal-to-blank (S/B) ratio when analyzing eight common drugs in whole blood.The most promising results came from formic acid and nitric acid treatments, which significantly enhanced analyte detection. Although these methods are currently time-consuming, Marwa envisions future industrial-scale production of pre-treated cartridges to support scalable clinical workflows.<h5>Key takeaways:</h5><ul><li>Substrate optimization can significantly reduce background noise in PS-MS analysis.</li><li>Acid-treated paper enhances S/B ratios, enabling more reliable detection of drugs in blood.</li><li>Industrial pre-treatment could make these improvements broadly accessible.</li></ul><h3>DLLME meets paper spray: improving sensitivity in plasma</h3>Saeed Almheiri’s presentation focused on dispersive liquid-liquid microextraction (DLLME) as a sample prep technique to increase sensitivity for PS-MS analysis of drugs in plasma, without adding significant complexity to the workflow.Using DLLME prior to VeriSpray analysis, Saeed demonstrated improved detection limits (as low as 0.3 ng/mL) across a panel of 10 drugs, including buprenorphine, cocaine, and MDMA. Importantly, this method maintained the fast, minimal-prep advantages that make paper spray ionization so appealing. Key takeaways:<ul><li>DLLME enhances analyte extraction from plasma, reducing matrix effects and improving quantitation.</li><li>Combining DLLME with VeriSpray maintains a low-complexity, high-speed workflow.</li></ul><h3>Conclusion: a new frontier for forensic and clinical toxicology</h3>The data shared at this workshop underscores the transformational potential of VeriSpray PaperSpray MS. She drives the implementation of ICH Q2(R2), Q14, and Analytical Quality by Design (AQbD) principles in collaboration with global biopharmaceutical partners. Amanda serves as a member of the United States Pharmacopeia (USP) Pharmaceutical Analytical Lifecycle and Data Science Expert Committee and is a board member of the ECA Analytical Quality Control Group. Before joining RIC, she spent more than 12 years at USP, where she worked both as a Senior Scientist – leading method development and validation projects, impurity analysis by LC-MS/GC-MS, and reference standards characterization by LC-HRMS – and as a Scientific Affairs Manager, contributing to major standards such as &lt;1220&gt; and &lt;1225&gt; and driving initiatives in AQbD, predictive modeling, and analytical procedure lifecycle management. Listeria monocytogenes, a foodborne pathogen, poses a particular risk to vulnerable populations such as infants, the elderly, immunocompromised individuals, and pregnant women. Timely and accurate identification helps to prevent outbreaks of listeriosis and is essential for ensuring food safety and protection of public health.
In this webinar, Dr. Helene Oberreuter, food microbiologist and senior government councilor, will highlight a validated workflow for the differentiation of Listeria monocytogenes serogroups by FT-IR spectroscopy. The workflow consists of species identification by MALDI-TOF mass spectrometry (MALDI Biotyper®) followed by serogroup differentiation with the IR Biotyper® using a classifier which is based on an artificial neural network.

Per- and poly-fluorinated substances (PFAS) are synthetic organic compounds widely used in products and processes because of their hydrophobic and non-stick properties. Known as ‘forever chemicals’ due to their persistence in the environment, PFAS are identified as a class of environmental contaminants of high concern. Increasingly, environmental and regulatory agencies are taking action to address PFAS contamination, setting sub-parts-per-billion (ppb) tolerance thresholds to limit exposure and protect public health.
While targeted techniques can detect these compounds at low levels, the number of current targets is limited. Consequently, the use of non-targeted High Resolution Mass Spectrometry (HRMS) is becoming important for the analysis of PFAS. This is due to the rapid rate of detection of novel PFAS in samples and a lack of analytical standards.

High Throughput Single-Cell Metabolomics

We present HT-SpaceM, a high-throughput method showcasing robust reproducibility in identifying metabolites in 10 cell lines.

Protein ubiquitination is a versatile post-translational modification that orchestrates many cellular pathways. Ubiquitin ligases are key specificity factors in this system. The HECT-type ubiquitin ligase HACE1 controls membrane dynamics and redox homeostasis by mechanisms that are still poorly understood at a structural level.
We determined the cryo-EM structures of HACE1 alone and in complex with its physiological substrate, RAC1. Together with biophysical analyses including HDX-MS and mass photometry, our studies reveal how HACE1 is regulated and how it recognizes its substrate with specificity. I am going to provide an overview of this work with a specific focus on the use of mass photometry.
