<h4>The science behind gummy candies</h4>Who can resist colourful, fruity gummy candies 🍬 with their signature chewy and elastic texture? While they may appear simple, their formulation is actually quite sophisticated. Each gummy is produced using a precisely balanced combination of gelling agents, sugars, acids, flavourings, and colourants, all working together to deliver the desired taste and texture.For instance, the characteristic chewiness of gummy sweets results from the interaction between gelatin and sugars. Even minor adjustments—such as changing the type or concentration of the gelling agent—can significantly alter the final product. In addition, manufacturers must meet regulatory and labeling requirements, including certifications like Halal compliance. This makes accurate and consistent monitoring of ingredients essential throughout production.<h4>The importance of SEC fingerprinting</h4>Size Exclusion Chromatography (SEC), also known as Gel Permeation Chromatography (GPC), offers a fast and reliable way to analyze gummy formulations. By separating molecules according to their hydrodynamic size, SEC generates a unique molecular “fingerprint” for each product.This fingerprint enables manufacturers to:<ul><li>confirm the type and origin of gelling agents (e.g., for Halal verification)</li><li>ensure batch-to-batch consistency and detect production variations</li><li>monitor changes during storage to assess shelf life</li><li>evaluate low-molecular-weight compounds such as sugars in a semi-quantitative manner</li></ul>Overall, SEC fingerprinting supports consistent product quality and regulatory compliance, ensuring that every gummy maintains the same expected characteristics.<h4>Influence of column selection on SEC performance</h4>Column selection plays a crucial role in achieving high-quality separations and detailed molecular fingerprints. To illustrate this, porcine and bovine gelatin standards were analyzed using two different configurations:<ul><li>a single Multipore <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/1655">SuperOH-P column</a></li><li>a coupled system of SuperOH-P 350 and SuperOH-P 150 columns</li></ul>Although both setups cover the same molecular weight range (100–1,000,000 Da), the coupled configuration effectively doubles the separation length. This results in improved resolution, which was confirmed experimentally. For example, combining a Refractive Index Detector (RID) with a Diode Array Detector (DAD) allows simultaneous detection of sugars, acids, and polymers within a single analysis.Accurate peak identification further enhances reliability. More details on the flow marker and its use in SEC are described in <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/VTN_0043_knauer_gpc_sec_data_reproducibility_with_flow_marker_96ee6b3cd9.pdf">this technical note</a>.<h4>Verification of gelatin origin using SEC</h4>Determining the source of gelatin is critical for proper labeling and compliance, particularly for Halal-certified products where porcine gelatin is not permitted.To demonstrate SEC’s capability, two gummy samples with identical formulations but different gelatin sources were analyzed. One sample contained porcine gelatin, while the other used bovine gelatin. The resulting SEC fingerprints showed clear differences in the gelatin region, allowing unambiguous identification. While SEC is not a replacement for fully validated HPLC methods, it serves as a rapid screening tool for comparing samples and supporting quality control.Using RID detection, differences in sugar composition become visible. Mass spectrometry imaging (MSI) began its development in 1997 with the publication of “Molecular Imaging of Biological Samples: Localization of Peptides and Proteins Using MALDI-TOF MS” by Richard Caprioli et al. in Analytical Chemistry. Over the past 25 years, there have been numerous developments in terms of samples preparation, matrix application, and instrumentation that have enabled increases in figures of merit related to MSI.
Each improvement is accompanied by an increase in method complexity and price tag, while many users are seeking an entry-level instrument to perform simpler experiments that still produce spatially resolved mass data.
To this end, we have tested a benchtop Shimadzu MALDI-8020 capable of MALDI MSI and employed simpler sample preparation techniques.
As an introduction to MSI, we share the optimization of relevant parameters and matrices as well as the results of relevant imaging experiments.

Single cell proteomics is growing as an analytical tool, delivering in-depth information of molecular processes occurring at the cellular level. The release of the timsTOF SCP in June 2021 opened the gate to label-free analysis of single cells.
This approach removes the needs of labelling and their associated costs and bias. Removing this step is not sufficient to handle all the challenges associated with the extremely diluted sample preparation, handling and processing.
In this live from the lab, we will present and demonstrate the use of a complete SCP ecosystem allowing to address the sensitivity and robustness issues at all stages of the analytical process, starting from single cell sorting and digestion down to the data analysis, considering sample transfer, separation, dia-PASEF® acquisition and real-time data processing.

Cell line development scientists and upstream bioprocess engineers aim at optimizing biotherapeutics production processes to maximize drug product quality, yield, and manufacturing efficiency. Currently, information on critical process and product attributes required for informed decision making is often insufficient and not readily available. This has a negative impact on the process robustness, throughput, and outcome.
Direct measurement of cell culture media and product quality attributes (PQAs) has recently been offered thanks to the Waters BioAccord™ LC-MS system with dedicated workflows for process analytics of biopharmaceuticals. The BioAccord™ LC-MS can be directly coupled to multi parallel bioreactor systems, like the Sartorius Ambr® 15 or 250 HT platforms, for accelerated process development. 
With this integrated solution, process engineers have direct access to the information they need to be in control: drug substance quality and yield can be maximized, process and product related impurities minimized, and more robust processes can be developed, thereby de-risking batch failure. This ultimately results in accelerated drug product development and manufacturing release from months or weeks to days, saving time and resources but more importantly enable faster delivery of safer treatments to patients.

He has published over 40 scientific papers and 50 posters in the areas of mass spectrometry, biomolecule characterization and bioseparations, and has been an invited presenter at national and international conferences on these topics. He is currently the Global Marketing Manager at Waters Corporation – Bioprocessing, where he is dedicated to deliver analytical solutions to advance upstream and downstream bioprocess.

Terence Teo has more than 15 years of experience in bioprocessing, specializing in downstream processing R&amp;D during the early phase of his career before moving to business development roles in GE Life Sciences (now Cytiva), Pall Biotech and most recently, Waters Corporation. Terence is motivated in supporting regional customers in APAC with technologies enabling continuous bioprocessing. He is currently working actively in Waters to drive the use of LC-MS analytical solutions to support near real-time CQA measurement in bioprocess applications, as well as use of LC-MS in multi-attribute method (MAM) and emerging drug modalities such as mRNA and viral vectors. Terence earned his M.Eng degree in Chemical and Biomolecular Engineering at National University of Singapore (NUS).

She has received her PhD from the University of Sydney (Chemistry) working on surface initiated polymerisation on silica nanoparticles using RAFT polymerisation and bioorthogonal conjugation of peptides using Native Chemical Ligation. She then spent 5 years working on developing platforms for stem cells and organoids characterization that included working with microfluidic devices and helical polymers. Naatasha joined Shimadzu in 2022 to expand her experiences to MALDI characterization on materials and biological samples.

Her recent work has focused on developing targeted metabolomics methods and routine Q-TOF workflows for both expert and non-expert users alike. Previously, Karen worked on forensic, metabolomics, and biomarker discovery applications with ambient ionization sources. She received her PhD in Analytical Chemistry from Purdue University where studied under the advisement of Prof. R. Graham Cooks.
