<div>Mulching films used in agriculture gradually break down and can become a significant source of microplastics in soils. A new study shows that these particles affect not only soil structure but also microbial life and nitrogen cycling.“We wanted to understand how microplastics from both conventional and biodegradable mulching films influence soil properties, microbial diversity and functions in real conditions,” explains Klára Šmídová, the study’s co-author . “That’s why we added realistic amounts of microplastics to soils in experimental plots in Finland, Germany, and Spain, monitored them over two growing seasons, and compared them with the control plots.”The results revealed that microplastics alter soil characteristics and microbial communities, although not always in the same way. “ However, a consistent finding was the decline in microbial activity associated with key processes, including nitrogen cycling, with negative effects increasing at higher microplastic concentrations and becoming more pronounced in the second year,” Šmídová adds. Because the microplastic levels used in the study reflect amounts already found in European soils, the findings highlight real risks to soil health. “We also found that the effects were stronger in southern than in northern Europe, likely due to differences in climate and soil conditions. This means that protecting soils from plastic pollution will require region-specific risk assessments,” she says.Over 60% of European soils are estimated to be unhealthy, with ongoing degradation driven by unsustainable land use, contamination, climate change, and extreme weather events. In response, the EU Parliament adopted the Soil Monitoring Law in October 2025, aiming to achieve healthy soils across Europe by 2050. “The law highlights the role of soil health in food security, water quality, and climate objectives, and establishes consistent soil monitoring across Member States. It includes measures to support farmers and foresters, while also creating a public list of potentially contaminated sites and an indicative watch list of emerging soil contaminants,” explains Klára Šmídová.The project brought together 19 research groups across Europe. “After each season, hundreds of samples were colle cted and sent to partners for analysis. This paper focuses on soil properties, microbial diversity, and greenhouse gas emissions. Two more manuscripts—on plastic additives and microplastic fate, and on soil invertebrates—are in preparation. This study investigates the effects of agricultural MPs from conventional (linear low-density polyethylene, PE) and biodegradable (starch-blended polybutylene adipate co-terephthalate, PBAT-BD) mulching films on soil physicochemical properties, aggregation, microbial diversity and functions, litter decomposition, and greenhouse gases emissions (GHG). For this purpose, MPs were mixed into soils at realistic MP concentrations of 0.005&nbsp;% and 0.05&nbsp;% (w/w) in 2022 on experimental plots in three EU countries representing different pedoclimatic conditions (Finland, Germany and Spain), followed by monitoring of the above-mentioned variables in the subsequent growing seasons 2022 and 2023. We found several significant MP-induced effects for soil properties, aggregation, microbial diversity, litter decomposition, and GHG, but the effect endpoints were less pronounced or varied considerably. Contrarily, microbial activity, contributing to soil functions such as nitrogen cycling, was consistently reduced by both conventional and biodegradable MPs. The reductions were more pronounced after the second season and for the higher MP treatment. As we aim to make our environment safer, it is important to detect contaminations at very low levels. Hundreds of pesticides are routinely used for crop protection across the globe and most of these pesticides can leach into our water systems. Therefore it is important to monitor these compounds down to very low levels in both environmental and drinking water. European regulations are under constant review, one good example is the proposed amendments to the Directive 2008/105/EC.
Given the challenges brought in by the new regulations, it is becoming ever so important to have sensitive and robust methodologies. The growing target list now include multiple pesticides groups such as pyrethroids, organochlorines and organophosphorus. 
Our recommendation for the GC amenable pesticides analysis is APGC. This technology allows analysts to reach lower levels than ever before. The APGC ionization is considered ‘soft‘ when compared with techniques such as EI, which means that less fragmentation is observed for many compounds. Reduced fragmentation simplifies pre-cursor ion selection, subsequently resulting in higher sensitivity and specificity in MS/MS analyses.

Trends and Challenges in PFAS Analysis

Survey findings and perspectives on the latest trends and best practices in PFAS analysis.

The development and expansion of analytical methods for per- and polyfluoroalkyl substances (PFAS) in food is essential for the continued monitoring of the United States (US) food supply and assessments of dietary intake.
In March of 2022, the European Union Reference Laboratory (EURL) for halogenated persistent organic pollutants (POPs) in Feed and Food released a guidance document covering priority PFAS of interest, including analytical method parameters and limits of quantification (LOQs). As a result, the Food and Drug Administration (FDA) began method extension work to incorporate ten new additional analytes to method C-010.02 including long chain perfluorosulfonic acids, fluorotelomer sulfonates, and perfluorooctane sulfonamide. In December of 2022, The European Union published Commission Regulation 2022/2388, establishing maximum levels for perfluorooctane sulfonic acid (PFOS), perfluorooctanoic acid (PFOA), perfluorononanoic acid (PFNA), and perfluorohexane sulfonic acid (PFHxS) in certain foodstuffs, primarily fish, molluscs, crustaceans, and eggs. As a result, the FDA method was evaluated for performance in reaching EU required LOQs for both compliance and monitoring purposes. The FDA method was found to be able to reach all required LOQs for analytes in matrices with established maximum levels and with further method modifications, required LOQs could be met in fruits, vegetables, and milk.

Understand the FDA’s approach for PFAS LOQs in various food matrices.

Glycosylation plays a crucial role in determining the pharmacological properties of biotherapeutics. The molecular heterogeneity of glycosylated biotherapeutics makes it difficult to provide thorough characterization of intact glycoproteins by mass spectrometry.
Current methods for analyzing biotherapeutics only measure fragments of the compound or rely on partial digestion of the glycans to reduce sample complexity. These limitations pose important risks and challenges for biotechnology companies that need to report human-compatible and consistent glycosylation, critical for drug safety and efficacy. 
We will present a new approach to glycoform fingerprinting that uses proton-transfer charge-reduction with gas-phase fractionation to analyze intact glycoproteins by mass spectrometry. The method provides a detailed landscape of the intact molecular weights present in biotherapeutic protein preparations in a single experiment and offers insights into glycoform composition when coupled with a suitable bioinformatic strategy. In addition, we highlight the application of charge detection native mass spectrometry (Direct Mass Technology mode) towards the elucidation of masses and aggregation of heterogeneous glycosylated biotherapeutics.

Pharma conclave 2023 (Day 2)

Developing targeted LC-MS/MS work to quantify key membrane receptors and antigens to evaluate drug targets and the effect of biotherapeutics is extremely challenging.
The expression levels of these proteins are often exceptionally low, the properties of these membrane proteins can limit the effectiveness of extraction and recovery, and the noises from biological matrices are typically high. Traditional methods often suffer from insufficient sensitivity and quantitative accuracy.
The focus of this webinar is to introduce an alternative method designed to implement more generic sample preparation steps in combination of strategic trap-elute chromatography, FAIMS selectivity, and sensitive LC-MS/MS acquisition utilizing selected reaction monitoring (SRM) on a premier triple quadrupole mass spectrometer. Prof. Qu and his team will demonstrate confident quantitation of various membrane receptors and antigens that are important to the biotherapeutics. The presented workflow will also demonstrate advantages to streamlining method development to increase productivity.

Jun Qu is a professor in the Department of Pharmaceutical Sciences of SUNY-Buffalo, and the director of Proteomics and Pharmaceutical Analysis Group in NY Center of Excellence in Life Sciences. His research is focused on the study of Clinical/Pharmaceutical Proteomics and Pharmaceutical Analysis by developing novel LC/MS-based strategies to markedly enhance the sensitivity, specificity, throughput and accuracy/precision for protein analysis in highly complex biological matrices. His research programs include i) highly sensitive and accurate characterization of biotherapeutics, targets and markers in pharmaceutical and clinical systems; ii) precise and large-scale global quantification of pathological proteomes using a novel MS1-based proteomic pipeline (IonStar); iii) sensitive identification, localization and quantification of post-translational modifications in tissue proteomes; iv) targeted investigation of marker proteins that are of high interests for clinical and pharmaceutical investigations. Qu lab is also one of the world leaders in LC-MS-based characterization of biotherapeutics in pre-clinical models. He has around 130 peer reviewed publications and &gt;70 grants funded as the principal investigator or co-investigator.

Mass Spectrometry Imaging (MSI) is a technique that allows chemical information to be obtained directly or via the use of matrices from a wide range of surfaces.
MS Imaging provides information on the spatial distribution of molecules at or near the surface. Imaging provides beautiful and information-rich images that allow scientists to answer the fundamental question: “Where is this component in my sample?”
In this webinar series, we will examine the basics of MS imaging, compare the techniques of untargeted and targeted MS Imaging and the information that can be gained from each, and end with an interview from Dr. Ryan Leib (Stanford University) and Dr. Roy Martin (Waters Corporation) on the practicalities of setting up a MS Imaging Core lab.

The steps necessary to set up an MS imaging core lab and answer questions such as: “who runs this type of lab?” and “who are the main customers?”.

Single-cell proteomics by mass spectrometry can provide valuable insights into distinct cell states and signaling patterns present in a cell population. However, carrying out proteomics profiling from the limited amount of material encapsulated in an individual cell presents significant challenges.
Tremendous efforts have been made to optimize all aspects of single-cell proteomics by mass spectrometry, with the aim of minimizing losses during sample preparation and maximizing sensitivity of data acquisition. Nevertheless, continuous development is needed to increase both the quantifiable proteome depth per cell, the quantitative accuracy and precision of those measurements, and the number of cells measured per unit time.
This webcast will present recently-developed approaches, ranging from sample preparation and chromatographic separation to advanced data acquisition, data interpretation and visualization methods. Advances in instrumentation, including the novel <a href="https://lcms.cz/products/836">Orbitrap Astral MS</a>, and how to use both TMTPro-labeled and label-free approaches, will be presented, showcasing the application of these methods to biomedical questions, and providing an overview of the current technological state of the field.
