Meeting EU Drinking Water Standards: The Critical Role of Sample Prep in Trace Contaminant Analysis

Although modern agriculture needs to be based on accurate real-time data on soil, plant, and environmental conditions, affordable, sufficiently sensitive, and robust monitoring devices for this purpose are currently missing. According to scientists, low-cost sensors based on new smart materials and printing technologies offer a suitable solution. The results of multidisciplinary research thus far and proposals for the development of these devices have been presented by researchers from Czechia, Great Britain, Spain, and the USA in a review article in the prestigious journal <a target="_blank" rel="noopener noreferrer" href="https://www.nature.com/articles/s41467-026-68778-6#Abs1">Nature Communications</a>.“Sensor technologies for monitoring plant health are still in their infancy. Only simple sensors capable of monitoring basic parameters such as pH, soil moisture, or temperature are commonly available on the market. However, if farmers are to face current challenges, including climate change, and ensure sufficient food for humanity, they need much more effective tools that will enable them to monitor, for example, various pathogens, including bacteria and fungi. Materials research and the use of graphene-based smart materials were led by scientists from CATRIN at Palacký University and VŠB-Technical University of Ostrava. Per- and polyfluoroalkyl substances (PFAS) are synthetic compounds that have been the focus of research development for targeted quantitation.
PFAS are present in a variety of consumer products as well as firefighting foam that collectively can be introduced into the environment. Due to the chemical structure, these compounds are hard to break down and are therefore labeled as forever chemicals. As such, once PFAS compounds have been introduced into the environment, they can leach from their origin into the environment and have been detected in surface waters, soils, dust, plant tissue, and even in the air. Recently, concerns have escalated not only for understanding the wider environmental impact, but animal and human health have evolved as research has now focused on detecting and quantifying in human plasma and tissue.
The focus of this webinar is to explore the range of sample types examined for routine PFAS detection and quantification. Dr. John Bowden will provide an overview of the challenges non-water matrices presents for extracting and quantifying including finding blanks and validating the workflows. In addition, research will cover new methods to introduce a wide range of PFAS onto UHPLC columns and perform low-level detection and quantification of PFAS in human plasma by LC-MS/MS. The second part of the webinar will focus on targeted PFAS quantitation using the Thermo Scientific™ TSQ Quantis™ Plus mass spectrometers supporting the EPA draft method 1633 which includes sample preparation using solid phase extraction (SPE) prior to LC-MS/MS analysis. <h4>IMMUNOPEPTIDOMICS ON THE EVOSEP ONE – NEW LEVELS OF SAMPLE THROUGHPUT AND PRODUCTIVITY</h4>
Immunopeptidomics is the study of peptide antigens that are bound to and presented on the cell surface by molecules encoded by the Major Histocompatibility, a highly polymorphic region of the genome that encode molecules involved in immune responses. These peptides are derived from both intracellular and extracellular degradation of the parental proteins, generating a highly complex repertoire of peptide ligands, including peptides derived from pathogens, tissue specific and cancer specific antigens when relevant. In humans, human leukocyte antigen (HLA) class I and class II molecules are of particular interest for immunopeptidomic analysis. These molecules are encoded by several loci and constitute thousands different alleles that vary between individuals. Thus, immunopeptidomes are highly individualized and almost impossible to accurately predict. This predicates the empirical measurement of the complexity and diversity of immunopeptidomes using mass spectrometry. Due to the relatively small amounts of material available for analysis the use of nanoflow LC-MS for immunopeptidome analysis is critical. Here I explore the implementation of the high throughput EvoSEP one platform couples to a Bruker timsTOF Pro 2 for robust analysis of immunopeptidomes across a variety of tissues and cell lines. Using the Whisper 20SPD method data quality and depth is maintained whilst sample carryover is negligible allowing more productive use of instrument time; I highlight significant improvements in throughput with over 1500 immunopeptidomes characterised in just 3 months.

<h4>MAPPS: High throughput immunopeptidomics for the assessment of the immunogenic potential of protein biotherapeutics</h4>
Biologics, mostly monoclonal antibodies and recombinant proteins, are a class of drugs used to treat some particularly difficult conditions. These proteins, when recognized as non-self by the immune system, can elicit unwanted immunogenicity. One of the first steps in this cascade is the presentation of fragments of this protein by MHC complexes on antigen presenting cells, which might ultimately lead to generation of anti-drug antibodies (ADAs). These ADAs can lower the therapeutic efficiency and can lead to severe immunogenic effects. To bring safe biotherapeutics to the market it is critical to thoroughly assess the immunogenic profile of a biotherapeutic. MAPPS (MHC associated peptides proteomics) assays are the method of choice to define all potential immunogenic regions of a protein. ImmuneSpec, an advanced immunopeptidomics platform, offers MAPPS as a service. Here we show how the combination of high-throughput MHC peptide purification, fast LC using EvoSep, and high sensitive MS addresses these needs.
