This flexibility is critical when working with limited or precious samples such as enriched cell surface glycoproteins, providing the sensitivity and robustness needed to characterize highly complex glycopeptides and intact glycoproteins.<h3>From forensics to glycoproteomics</h3>Riley’s scientific path began in undergraduate chemistry and psychology, where an early interest in forensics led him to mass spectrometry. Rather than focusing on applications, he became interested in the instruments themselves.That fascination took him to the University of Wisconsin-Madison, where he trained in Josh Coon’s lab and worked extensively on electron transfer dissociation. While developing new fragmentation methods, Riley encountered glycopeptides almost by accident and quickly realized their complexity and importance.A postdoctoral position in Carolyn Bertozzi’s lab at Stanford deepened his exposure to glycobiology and reinforced his interest in building tools that allow scientists to better interrogate biological systems. That philosophy now defines his research program at the University of Washington.<h3>Why flexibility matters in glycoproteomics</h3>Glycoproteomics presents unique analytical challenges. Glycans increase molecular heterogeneity, complicate ionization, and require multiple fragmentation strategies to fully characterize.“There’s no single method that can answer all of our questions,” Riley explains. “We need flexibility at every level.”For the Riley Group, that flexibility comes from advanced LC-MS platforms, particularly Tribrid instruments like the Orbitrap Ascend Mass Spectrometer. These systems allow his team to combine ETD, HCD, PTCR, and high-mass analysis within a single workflow.That versatility is essential when working with limited samples, especially in experiments involving cell surface glycoproteins or double enrichment strategies.<h3>Turning technology into biological insight</h3>One recent study highlights how instrumentation directly enables discovery. Riley’s team investigated how glycosylation affects protein corona formation using SEER nanoparticles. The workflow required two enrichment steps, leaving very little material for analysis.Using the Orbitrap Ascend Mass Spectrometer’s real-time library search capabilities, the team developed Autonomous Dissociation Type Selection, a method that uses real-time decision making to determine the optimal fragmentation method during acquisition. This approach allowed the team to extract comprehensive information from a single LC-MS injection, preserving sample while accelerating discovery.“That kind of experiment simply wouldn’t be possible without real-time decision making on the instrument,” says Riley.<img src="https://storage.googleapis.com/labrulez-bucket-strapi-h3hsga3/Thermo_Fisher_Scientific_What_Happens_When_Glycoproteomics_Meets_Real_Time_LC_MS_Decision_Making_2_f5c7de51c3.jpeg" alt="Thermo Fisher Scientific: What Happens When Glycoproteomics Meets Real-Time LC-MS Decision Making?: Kayla Markuson, B.S., is one of the grad students in the Riley Group. Students in Riley’s lab are encouraged to design experiments around unanswered biological questions rather than technical constraints.“When the instrument isn’t the bottleneck, creativity becomes the skill you’re really teaching,” he notes. “That’s how you prepare scientists for the future.”<h3>Looking ahead</h3>As instrumentation continues to evolve, Riley believes the next breakthroughs will come from increased sensitivity, richer fragmentation strategies, and smarter data analysis. The ultimate goal is not just better measurements, but deeper biological understanding.“Glycoproteomics demands multiple analytical strategies,” Riley explains. “We need to move seamlessly between different fragmentation methods, mass ranges, and separation conditions. Light scattering techniques deliver multi-attribute quantification (MAQ) of mRNA and lipid nanoparticle-formulated mRNA and can play a key role in product and process development as well as quality control of mRNA-LNP therapeutics. Specifically, multi-angle light scattering (MALS) coupled to separation by field-flow fractionation (FFF) or size-exclusion chromatography (SEC) provides high-resolution quantification of several mRNA and LNP attributes including size distribution, physical stability, LNP number concentration, morphology, and mRNA and lipid concentration.
The method can also be applied to evaluate how these attributes are affected by preparation, formulation, and storage conditions. In addition to high-resolution MAQ by SEC/FFF-MALS, dynamic light scattering (DLS) and electrophoretic light scattering (ELS) can be used for rapid, automatable batch measurements of particle size and concentration, polydispersity, and zeta potential, enabling rapid screening of the impact of different ions, ionic strength, pH, temperature and other parameters on the biophysical properties and stability of mRNA and LNP-mRNA.

AAVs are attractive vehicles for targeted drug delivery. There are numerous Critical Quality Attributes (CQAs) evaluated to ensure safety and efficacy of AAVs. Aggregation is a CQA and should be controlled as AAVs are prone to aggregation from freeze thaw cycles during storage.
<ul>
<li>In this webinar, we will explore CQAs using a novel size exclusion column optimal for AAV analysis, which enables the separation of AAV monomer from aggregate species with excellent peak shape, resolution and peak profile to provide better data quality. Due to both the inert surface chemistry and flow-path in the BiozenTM dSEC-7 column, the analysis conditions were independent of the serotype showcasing the utilization of platform methodology.</li>
<li>In addition to calculating the aggregation level, aggregate molar mass, molar masses of protein and nucleic acid, and extinction coefficients of capsid and DNA were also determined. The novel particle chemistry of Biozen dSEC-7 provides instant stabilization of sensitive detectors like MALS to provide quality data.</li>
<li>Additionally, Full-to-Empty capsid ratio was determined to accurately predict capsid load demonstrating Biozen dSEC-7’s utility as a holistic solution for the determination of CQAs of AAVs.</li>

Lipid nanoparticles (LNPs) are crucial for RNA therapeutics, notably after the success of Patisiran and COVID-19 mRNA vaccines. LNPs need precise design for effective clinical use, but the understanding on mixing methods influences the LNP structure and biological activity is limited.
This study addresses this gap by comparing standard and advanced characterization methods including multi-wavelength analytical ultracentrifugation (MWL-AUC), multi-angle light scattering (MALS), and synchrotron size-exclusion chromatography small-angle X-ray scattering (SEC-SAXS) with singular-value decomposition methods (SVD). We will present the strengths and weaknesses of each approach and showcase the increased detail newer advanced methods provide by comparing LNP formulations made using two common small-scale production methods: microfluidic rapid mixing and bulk mixing. The characterization techniques employed here can enhance our understanding of LNP structure-function relationships and enable researchers to define their RNA LNP products more precisely, which can improve LNP quality and potentially accelerate pharmaceutical development.
