This was an expected result of EFSA’s letter of concern on those 4 PFAS and labs reacted accordingly. A more challenging task came with the EURL PFAS Guidance Document and a bit later with the Recommendation EU 2022/1431. The target LOQs for the EU 4 PFAS were set at 0.001 µg/kg in fruits, vegetables and babyfood. As regulations, like the EU Drinking Water Directive, the EU Groundwater Directive, and the EU Water Framework Directive, have been updated or are in the process for an update, the requirements for method sensitivity have become more demanding. Whilst concentration levels go down, direct injection methods are increasingly desirable to laboratories carrying out routine analysis of PFAS where analyte preconcentration is not deemed necessary or required. They have a simple sample preparation method, which not only reduces the opportunity for contamination and error but speeds up analysis time and the requirements for large volumes of samples. PFAS Analysis in Drinking, Ground, and Surface Water

We will give insights how the analysis of PFAS in environmental samples works in routine testing labs.

We developed and validated multiple novel diagnostic methods. First, an immuno-affinity liquid chromatography–tandem mass spectrometry (ILM) assay that quantifies Mycobacterium tuberculosis CFP10 peptides and HIV-1 p24 in serum, showing high sensitivity and specificity in both adults and children. This method allows early detection of HIV and TB and monitors treatment responses effectively. The second approach involves an automated peptidomics pipeline that rapidly identifies mycobacterial species and subspecies from early growth cultures, including drug-resistant strains, significantly reducing diagnosis time compared to current clinical practices. MassHunter Acquisition for LC/TQ and MRM Method Optimisation

This session will provide a comprehensive overview of the various aspects of MassHunter Acquisition for Liquid Chromatography/Triple Quadrupole (LC/TQ) systems.

<p><strong>Measurement of FT4 and FT3 is typically performed by immunoassay, however these can be prone to suffer from interferences and a lack of selectivity. Currently, the accepted gold standard for the measurement of FT4 and FT3 is using equilibrium dialysis (ED) and combined with the use of LC-MS/MS can overcome the issues with immunoassay in the measurement of FT4 and FT3. However, the equilibrium dialysis procedure can be quite time consuming and often being performed overnight.</strong></p><p>Here, we describe a simple clinical research method to perform ED using a commercially available device followed by sample preparation by solid phase extraction (SPE) and UPLC-MS/MS analysis using the Waters™ ACQUITY™ UPLC and analytically sensitive Xevo™ TQ Absolute Mass Spectrometer for clinical research. Data analysis and review has been simplified with the use of the waters_connect™ with QUAN Review Software. This has allowed for the complete workflow including equilibrium dialysis to be performed within a typical working day.</p><p>Simplified Equilibrium Dialysis for the UPLC-MS/MS Analysis of Free Testosterone (FT) for Clinical Research</p><p>Historically, the measurement of free testosterone in human serum has presented a challenge for laboratory scientists, with such a large portion bound and the complexity and time involved in “gold standard” equilibrium dialysis. Bioanalytical assessment of unbound compounds in blood, brain tissue, and cerebrospinal fluid describes its distribution for the systemic circulation into the CNS. The inherently high sensitivity and specificity of LC-Triple Quadrupole Mass Spectrometry enables low level detection in small sample volumes, allowing serial sampling from single animals, reducing animal use and inter-animal variability, supporting the 3Rs principles (replacement, reduction, refinement)</span></p><p><span name="summary">The bioanalysis of neurotransmitters such acetylcholine and monoamines, is challenging due to low molecular weight, high polarity, instability, and low endogenous concentrations. At Sygnature, the </span><i><span name="summary">in vivo</span></i><span name="summary"> Pharmacology team employ microdialysis to permit assessment of neurotransmitter changes in specific brain regions.
