These techniques are often employed as initial purification or concentration steps before further refinement.<h5>Precipitation Techniques</h5>Protein precipitation is a widely used approach in purification workflows, typically relying on one of three main methods:<ul><li>Organic solvent precipitation involves adding solvents like ethanol or acetone to lower the solution's dielectric constant. This promotes protein–protein interactions, reducing solubility and leading to aggregation and precipitation.</li><li>Isoelectric point precipitation exploits the pH at which a protein carries no net charge. At this point, solubility is minimized, causing the protein to precipitate from solution.</li><li>Nonionic polymer precipitation uses polymers such as dextran, polyethylene glycol (PEG), or NPEP to alter the solution’s physical properties. In recent years, the use of quantitative liquid chromatography (LC) coupled charged aerosol detection (CAD) for poor UV absorbing analytes in multicomponent mixtures has grown exponentially across industrial sectors.
In this live webinar, we will highlight some distinctions between CAD and other UV-absorbance detection, such as the non-linear response to analyte concentration, and sensitivity to mobile phase impurities. However, careful optimization of CAD parameters can lead to successful development and transfer of analytical methods, nearly on par with that of conventional UV absorbance detectors. Through careful tuning CAD parameters, we have developed quantitative LC-CAD assays which are sensitive, precise, accurate and robust across early and late-stage pharmaceutical development. Examples of LC-CAD methods across various stages of drug development spanning from discovery to commercialization will be shared during the presentation.

The scientific community aims to enhance transparency and reliability in published research, with a specific focus on the quality of biological reagents, notably recombinant proteins. Concerns have been raised regarding irreproducible results, prompting discussions on common quality issues associated with recombinant proteins and their impact on downstream applications. In response, professionals in protein purification and characterization have collaboratively developed guidelines for improved quality control.
These guidelines include minimal information requirements for protein identity, production parameters, and long-term stability, as well as a minimal set of quality tests for purity, homogeneity, and identity. Additional recommendations cover DNA contamination, “spectral and thermal denaturation signatures”, homogeneity, “competent fraction”, storage conditions, batch-to-batch reproducibility, and other factors based on intended applications. Evaluation of these guidelines over a one-year period suggests that their implementation can enhance experimental reliability and optimize protein quality. Investing in protein QC benefits all stakeholders in life sciences, including researchers, editors, and funding agencies, by improving data veracity and minimizing resource wastage.

Characterizing the ratio of tumor-killing payload to cancer-binding antibody (DAR) is a critical quality attribute needed to determine the potential potency of a given ADC candidate. This webinar features HIC as a solution for quantifying DARs in ADC biotherapeutics. Following that, Dr. Christoph Bauer, Associate Principal Scientist, will discuss the IT architecture and integration aspects of the solution implementation.A lab with significant analytical capacity entails managing a large number of instruments and extensive analytical data on a daily basis. Within our purification labs, analytical screening data generated for compounds submitted for purification, as well as data from instrument suitability testing, was traditionally reviewed manually by an analyst to determine the most appropriate method for purification. However, this changed with the implementation of the automation solution from Mnova. The solution includes the Chrom SST brick, which monitors the performance of our instruments, and Chrom Best Method, which automatically processes the screening data and identifies the best conditions for purification, thereby freeing up analyst time.In this webinar, we will explain how these solutions fit into our day-to-day work and the benefits they have brought us.
