This generates radical species that can undergo new types of reactivity.</p><p>Repka explains that one of the key advantages of photoredox chemistry is its ability to operate under mild conditions, often using visible light. This makes it attractive for sensitive molecules and biological systems, including potential applications in drug release, live-cell chemistry, and drug development.</p><h4><strong>Photocrosslinking and drug-target interactions</strong></h4><p>Repka’s group initially aimed to develop visible-light photocrosslinking methods to study drug interactions with biological targets. Photocrosslinking can covalently link a drug to its target, making it easier to identify the biological target of a compound.</p><p>Traditional photocrosslinking often uses UV light, but visible light offers milder and more biologically compatible conditions.</p><h4><strong>From photocrosslinking to solvent activation</strong></h4><p>The project shifted direction when the desired photocrosslinking reactions gave very low yields. Instead of reacting with amino acids as intended, the radical intermediate mainly reacted with itself.</p><p>The team explored different solvents to stabilize the radical, but found that some solvents, such as DMF, were not inert. Instead, the solvent reacted with the photocatalyst or radical species, revealing unexpected solvent activation chemistry.</p><h4><strong>Why LC-MS was critical</strong></h4><p>LC-MS became essential because the reaction mixtures were too complex and low-yielding for NMR or GC-MS. High-resolution LC-MS allowed the team to detect minor products, identify photocatalyst modifications, and observe solvent addition products with high confidence.</p><p>Repka emphasizes that LC-MS was both a qualitative and quantitative tool. It was used quantitatively to measure how much photocatalyst remained after light exposure, and qualitatively to identify degradation products and new reactivity pathways.</p><h4><strong>Photocatalyst stability workflow</strong></h4><p>The team developed a systematic workflow to evaluate photocatalyst stability under light exposure. Because catalyst degradation was time-sensitive, reproducibility required strict control of experimental conditions.</p><p>Key steps included using a glove box, commercial deoxygenated solvents, accurate weighing on a microbalance, controlled light exposure in a photoreactor, and temperature control with a chiller. These precautions helped reduce day-to-day variability and enabled reproducible measurements of catalyst remaining.</p><h4><strong>Quantifying catalyst remaining by LC-MS</strong></h4><p>Calibration curves were prepared using pure photocatalyst solutions and LC-MS ion areas. The team worked in the nanomolar range to maintain linearity.</p><p>Instead of relying only on total ion chromatograms, they used extracted ion chromatograms (EICs) to improve signal-to-noise for the photocatalyst signal. Baseline subtraction was also applied to reduce noise and improve quantitation at low concentrations.</p><p>Calibration curves and quality control samples were prepared and measured on the same day as the reaction samples, because LC-MS signal intensity can vary significantly from day to day.</p><h4><strong>Why GC-MS was used later</strong></h4><p>For demethylation studies, the group moved to GC-MS because the substrates and products were small and volatile enough for gas chromatography. GC-MS was better suited for rapidly screening many reaction conditions, including different photocatalysts, solvents, and oxidants.</p><p>Instead of generating full calibration curves for every experiment, the team used an internal standard to streamline analysis and determine percent conversion based on relative response factors.</p><h4><strong>Practical lessons from the project</strong></h4><p>Repka highlights the importance of careful sample preparation, especially when working with very small quantities of photocatalysts. A high-quality microbalance, stable granite platform, antistatic tools, and aluminum weigh boats improved weighing accuracy and reproducibility.</p><p>She also argues that mass spectrometry should be a standard part of photoredox method development because it can reveal minor products, support reaction optimization, and provide insights that may be difficult or impossible to obtain by NMR alone.</p><h4><strong>Final perspective</strong></h4><p>Beyond the chemistry, Repka emphasizes the importance of student training and mentoring. <p><strong>The increasing popularity of extra virgin olive oil (EVOO) and the increasing problem of food fraud have provided the need for quality and authenticity control. Typical problems are the mislabeling of protected designation of origin (PDO) or edible oil adulteration. In this presentation we give an overview about the capabilities of different mass spectrometric methods to accurately determine the PDO of EVOO.</strong></p>

<li>Multi-class LC-MS and GC-MS methodologies together with statistical methods enable the discrimination between different PDOs, identifying potential origin markers.</li>
<li>The combined use of non-targeted and targeted approaches enhances the outcome of the study. GC-APCI-QTOF preserves the pseudo-molecular ion information, which is a great advantage and facilitates the identification.</li>
<li>Co-eluting isomers maybe PDO markers and these can be identified and characterized by timsTOF technology.</li>
<li>Rapid LC-free, magnetic resonance mass spectrometry (MRMS) analysis was used for mapping and quality control assessment of EVOO using statistical methods which allows for clustering according to geographical origin, harvesting year, cultivation practice and the oil production procedure.</li>

<p>Food fraud is increasingly common and includes activities such as misbranding, mislabeling, dilution, counterfeiting and adulteration. This presentation will describe a comprehensive approach that can be used for routine detection of food adulteration using classification model, built from “pure” (unadulterated) samples. The workflow can utilize either a high-resolution GC/MS or LC/MS, providing a flexibility of choice depending on sample matrix and/or instrument availability. We will discuss how to build a robust classification model in Mass Profiler Professional and how to deploy it for routine testing using Classifier software. Results from spices, fruit, and tea will be shared.</p>

<p><strong>Nitrosamine (genotoxic) impurities have been found in some angiotensin II receptor blocker (ARB) medicines, such as valsartan, and more recently in histamine-2 receptor blocker medicines, such as ranitidine, drawing close scrutiny from FDA, the European Medicines Agency (EMA) and other regulatory bodies leading to product recalls.</strong></p>
<p>Gas chromatography–mass spectrometry (GC-MS) has typically been the detection technique of choice; however, not all nitrosamines can be analyzed this way. As the number of known nitrosamines requiring trace level quantitation increases, the greater the need for robust, sensitive, and high-throughput assays to provide a cost-sensitive approach of a single analysis capable of accurately measuring all compounds. The use of liquid chromatography with high-resolution MS (LC-HRMS) allows low level detection and high confidence of a wider range of nitrosamine impurities.</p>

<p><strong>High-resolution, accurate-mass (HRAM) mass spectrometers (MS) are a class of MS instrumentation with capability to resolve complex sample matrix and to allow identification of compounds by measuring their accurate masses.</strong></p>
<p>In particular, those coupled to liquid chromatograph (HRAM LC/MS) have been used extensively for structural elucidation of unknown compounds primarily in chemical industry (impurity analysis) and biological researches (proteomics, metabolomics, lipidomics), but also increasingly in forensic science, food safety and environmental testing. Given the recent reduction of instrumentational cost, it will not take many more years until HRAM analysis becomes a versatile tool for all industries and researches. </p>
<p>The key performance attribute underlying all application fields is mass measurement accuracy (MMA) as it affords molecular specificity and reduction of false positive results. MMA is given in the specification of HRAM instruments in parts-per-million (ppm), frequently stating a result of a set of representative measurements e.g. “&lt;1 ppm RMS”. However, this by itself does not convey how repeatably and reproducibly this MMA can be achieved. </p>
<p>The webinar describes various metrics that are relevant to statistical interpretation of MMA, which explain why mass resolution of 30,000 is more than sufficient for mass measurement at sub-ppm accuracy. Using the Shimadzu LCMS-9030 quadrupole time-of-flight MS, the rationale and strength of instrument stability for consistently achieving sub-ppm MMA has been demonstrated in small molecules.</p>

Recently, several manufactured lots of Angiotensin II Receptor Blockers (Sartan) and Ranitidine drugs have been recalled due to the presence of high potency mutagenic impurities (nitrosamines) found in these products as a result of a manufacturing process change.</strong></p>
