Exposure to this class of chemicals is widespread and their ability to move and persist in the environment makes them difficult to clean up. Found in everyday household and workplace products such as drinking water, food packaging, fish caught from compromised environments, and personal care products is an indication of how widespread the use of PFAS and its contamination in the&nbsp;environment.&nbsp;</p><p>Traditional techniques of analysis to enhance sensitivity incorporate the use of solid phase extraction, which allows enrichment of the sample before injection. Increased awareness of emerging contaminants has necessitated the need for higher throughput methods, some bypassing the sample preparation step, to decrease sample analysis time. The calculated concentration was prior to dilution of methanol and demonstrates that all but three analytes were equal or less than 1 ng/L detection limits, the analytes with greater than 1&nbsp;ng/L MDL were 3:3 FTCA and 8:2 FTSA (Table 6).</p><h4><strong>Conclusion&nbsp;</strong></h4><p>The increased awareness of PFAS globally has meant that regulations are constantly being adopted and larger amounts of samples at lower limits are required. By incorporating the direct injection technique on the Agilent 6495D LC/MS we get a combination of high sample throughput while providing excellent analytical sensitivity. Also, increasing throughput and reducing consumables cost creates increased value in the instrument. The method displays a clean baseline and sensitivity with the observation of 1 ng/L detection in spiked wastewater. Reproducibility and robustness were measured between the concentrations of 0.5 to 100 ng/L on the 6495D LC/MS. The system was able to detect 38 PFAS between 0.25 to 3.4 ng/L with RSD &lt; 20% over 150&nbsp;direct injections, providing confidence in the data for a complex matrix like&nbsp;wastewater.</p><h3><strong>2. Shimadzu: A sustainable analytical approach for detecting extra virgin olive oil adulteration using Nexera™ UC</strong></h3><ul><li>Technical note</li><li><a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/c190_e341_c488032e81.pdf"><strong>Full PDF for download</strong></a></li></ul><p>Knowledge of the composition of triacylglycerols (TAGs) in vegetable oils is important for dietary and nutritional reasons, due to the influence of the different properties of each fatty acid on the human organism. Specifically, extravirgin olive oil (EVOO) is a popular food ingredient worldwide, and for this reason, detecting economically motivated adulteration is important to protect consumers’ interest and health[1]. According to the latest report from the EU Food Fraud Network (European Union, 2021), olive oil (OO) tops the list of reported adulterated food products. The official methods to assess OO purity and detect the presence of extraneous vegetable oils include the analysis of fatty acids (FAs), triacylglycerols (TAGs) and sterols (Regulation (EU) No 2568/91 and its amendments) [2].&nbsp;</p><p>A variety of chromatographic techniques has been employed for TAGs analysis and separation. High-performance liquid chromatography (HPLC) methods, which use reversed-phase (RP) and silver-ion (Ag) columns for TAGs separation, have been employed either independently or in combination[3]. However, both these approaches have the drawback of relying on toxic organic solvents. The alternative separation technique employed in TAGs analysis is supercritical fluid chromatography (SFC) coupled with a UV detector at very low wavelengths, due to the weak UV absorption of the mobile phase. In this study, a fast, simple and green methodology was optimized to detect intentionally adulterated olive oil with cheaper seed oils at different levels by means of SFC with UV detection, followed by statistical analysis.</p><h4><strong>2. Experimental</strong></h4><h5><strong>2.2 SFC-PDA Nexera™ UC system&nbsp;</strong></h5><p>All experiments were performed on a <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/library?mainauthorItems=1&amp;search=Nexera%E2%84%A2+UC"><strong>Nexera™ UC system</strong></a> (Shimadzu Europa, Germany). The system was equipped with a CBM-20A communication bus module, a DGU-20A5R degasser, an LC-30ADSF CO2 pump, one LC-20ADXR dual plunger parallelflow pump, a Sil-30AC autosampler, a CTO-20AC column oven, a SPD-M20A photodiode array (PDA) detector, and a SFC-30A backpressure regulator (BPR). Chromatograms were recorded using Shimadzu <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/library?mainauthorItems=1&amp;search=LabSolutions"><strong>LabSolution</strong></a> ver. 5.80 software. Four Ascentis Express C18 columns, provided by KGaA (Darmstadt, Germany), were connected in series for TAGs separation. The analytical conditions used are summarized in Table 1.</p><h4><strong>3. Results and discussion&nbsp;</strong></h4><p>Using four C18 columns packed with superficially porous particles, a highly efficient separation of TAGs with an isocratic elution mode was provided, allowed for complete sample TAG elution within 30 min with the conditions used in this method (Figure 1). Peak identification was carried out according to partition number (PN), the published composition of these common vegetable oils and the relative amount of major and minor peaks, and finally taking into account TAGs retention times. For compounds with identical PN, separation was achieved based on the number of unsaturations. The elution order of TAGs was primarily governed by the hydrophobicity of the solutes. As the PN increases, the non-polar character of TAGs becomes more dominant.</p><h4><strong>Conclusions&nbsp;</strong></h4><p>The Nexera™ UC system provided excellent results for assessing TAGs composition in ten different vegetable oils examined in this study employing an SFC-PDA analytical method using isocratic mode and UV detection at 205 nm. This approach does not require any sample preparation and enables analyses to be completed with less waste and less cost compared to traditional methods, making it feasible for routine analyses, while providing reliable quantitative values.&nbsp;</p><p>Quantification of adulterated EVOO was performed by establishing curves based on peak areas of LLL marker versus seed oils concentrations. Statistical analysis using PCA and one-way ANOVA also visually distinguished adulterated EVOO from pure EVOO, and the result was in accordance with the present quantitative method. This fast and environmental method exhibits a huge potential for quality control and authenticity evaluation of EVOO.</p><h3><strong>3. Thermo Fisher Scientific / ASMS: Uncovering biological differences at scale: high-throughput and in-depth plasma proteomics with the Seer Proteograph ONE workflow and Orbitrap Astral Zoom Mass Spectrometer</strong></h3><ul><li>Poster</li><li><a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/po_393_uncovering_biological_astral_zoom_seer_po393_asms2025_na_en_f697aef9c0.pdf"><strong>Full PDF for download</strong></a></li></ul><p>Plasma proteomics enables minimally invasive biomarker discovery by capturing disease-related molecular signatures. However, its effectiveness is limited by the wide dynamic range of protein concentrations. To address this, we combined the Seer Proteograph ONE workflow with the Orbitrap Astral Zoom mass spectrometer to enable a high-throughput, high-precision platform that delivers deep plasma profiling and actionable insights for large-scale clinical and translational research (Figure 1).</p><h4><strong>Methods:&nbsp;</strong></h4><p>The Seer Proteograph ONE workflow was used to prepare plasma samples from a small disease cohort with matched healthy controls to evaluate its performance and feasibility in detecting biologically meaningful differences. Samples were then separated and analyzed using the <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/398"><strong>Thermo Scientific Vanquish Neo UHPLC system </strong></a>coupled to the <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/1959"><strong>Orbitrap Astral Zoom mass spectrometer</strong></a>, enabling deep and reproducible proteome coverage. Two analytical approaches—a high-throughput method and a Max-ID workflow—were tested to demonstrate the versatility and throughput for comprehensive biomarker discovery in clinical cohort studies.</p><h4><strong>Results:&nbsp;</strong></h4><p>The Seer Proteograph ONE workflow with Orbitrap Astral Zoom mass spectrometer demonstrated that both high-throughput and Max-ID workflow were able to detect differences in biology in diseased states from healthy controls. The high-throughput workflow enabled rapid, reproducible analysis, while the Max-ID approach delivered deeper proteome coverage. The synergy between the Seer Proteograph workflow and Orbitrap Astral Zoom mass spectrometer supports scalable, high-resolution biomarker discovery, making it suitable for large clinical studies and precision medicine applications.</p><h4><strong>Conclusions&nbsp;</strong><span><strong>&nbsp;</strong></span></h4><ul><li>The Seer Proteograph ONE workflow combined with Orbitrap Astral Zoom mass spectrometer identified &gt;10,800 protein groups using ultradeep profiling, compared to &gt;8,200 with the high-throughput method—demonstrating scalable, reproducible plasma proteomics with clear detection of biological differences for biomarker discovery.&nbsp;</li><li>The Protoeograph ONE workflow combined with the Orbitrap Astral Zoom Mass Spectrometer captures exceptional dynamic range of 7-8 orders of magnitude in plasma proteome&nbsp;</li><li>&gt;1,000 differentially abundant proteins detected with biologically relevant dysregulated pathways such as immune signaling or tissue remodeling underscores the complex biology of lung cancer&nbsp;</li><li>Identification of FDA markers as differentially expressed showcases the workflow’s strength for capturing biologically relevant plasma proteins</li></ul><h3><strong>4. Waters Corporation: Improving Throughput and Analytical Greenness in Pharmaceutical Discovery Using Ultrashort (2.1 x 10 mm) HPLC Columns</strong></h3><ul><li>Application note</li><li><a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/720009074_en_406ea811f9.pdf"><strong>Full PDF for download</strong></a></li></ul><h5><strong>Benefits&nbsp;</strong></h5><ul><li>Up to 60% decrease in analysis time using the ultrashort (2.1 x 10 mm) column</li><li>Lower AMGS metric values indicating improvements in analytical method greenness</li></ul><p>Drug discovery is a fast-paced, high-throughput environment where chemists screen large numbers of samples to determine if a suitable compound has been created. Supporting these efforts from an analytical standpoint using liquid chromatography (LC) or liquid chromatography-mass spectrometry (LC-MS) can present challenges. For many discovery workflows, hundreds if not thousands of samples are analyzed for the determination of drug concentrations in biological matrices, for structural confirmation or for an assessment of purity. Analytically speaking, this means that the number of samples that may be analyzed per unit time, or throughput, is valued more highly than is achieving baseline separation of all components. To maximize throughput, rapid (“ballistic”) gradients can be employed along with short analytical columns.1 Ballistic gradients are designed to run at the highest flow rate possible while using the steepest gradient slope, in terms of percent solvent per column volume (%/VC).2 This allows for the fastest elution off the column possible. Shorter columns are used not only to limit the pressure generated by the column, thereby allowing faster flow rates, but also to reduce the analysis time.&nbsp;</p><p>In this application note, the use of a 2.1 x 10 mm 2.5 µm XSelect™ HSS T3 Column is examined compared to 2.1 x 30 mm and 2.1 x 50 mm columns packed with the same stationary phase. These three columns were used to analyze a full 96-well QuanRecovery™ Plate to simulate the output of a drug discovery environment where multiple samples need to be run as fast as possible. Prior to sample analysis, instrument methods were created to increase throughput by limiting the inter-injection delay. The inter-injection delay time was calculated and recorded. The total analysis time for each column configuration was determined and compared, along with the AMGS. While the loss of efficiency in using a 10 mm column was expected, the shorter column configuration achieved adequate results for a single component sample. For Waters™ LC systems, there are a few parameters in the inlet method that can be adjusted to speed up the time between injections, or inter-injection delay. This delay comes from the sequence of steps that the system takes to draw the sample and inject it into the flow path of the LC. The parameters that need adjustment are all located in the sample manager panel of the instrument method or inlet method. Figure 1 shows screenshots of the sample manager panel and where the mentioned parameters are located.</p><h4><strong>Conclusion&nbsp;</strong></h4><p>The use of high-throughput LC separations in pharmaceutical discovery allows new lead compounds to be selected quickly and efficiently. This may be achieved using short columns with ballistic gradients. To support these activities 2.1 x 10 mm columns were developed and used to analyze a full 96-well plate of samples. Comparing the results obtained on the 10 mm column to those produced using 30 and 50 mm columns showed a 57–146% higher throughput. Additionally, the use of the 10 mm column was determined to be substantially “greener” as measured by the analytical method greenness scores, with decreases of 40 and 68% relative to 30 and 50 mm length columns.</p>

Agilent Technologies: Large-Volume Dilute-and-Shoot Analysis of PFAS in Drinking Water Using the Altura PFAS Column</strong></h3><ul><li>Application note</li><li><a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/an_analysis_pfas_drinking_water_altura_5994_9186en_agilent_a66334e5b0.pdf"><strong>Full PDF for download</strong></a></li></ul><p>Per‑ and polyfluoroalkyl substances (PFAS) are synthetic fluorinated compounds valued for their oil‑ and water‑repellent properties, but their environmental persistence and mobility have driven the development of increasingly stringent regulations for drinking water monitoring. Existing dilute-and-shoot methods, such as EPA 8327 and ASTM D7979, were originally designed for screening PFAS in nonpotable water. These methods typically rely on 10 to 30&nbsp;µL injection volumes, setting practical quantitation limits appropriately 10 ng/L.<sup>1,2&nbsp;</sup></p><p>We present a large‑volume injection workflow that extends the capabilities of these methods toward trace PFAS quantitation in drinking water. Using the Agilent 6495D triple quadrupole LC/MS (LC/TQ) coupled with a 1290 Infinity II LC, we achieved injections of 200 µL of methanol‑diluted water on the Altura PFAS column without requiring SPE concentration. The Altura PFAS column maintained peak symmetry and retention stability under these injection conditions. This configuration enabled a working calibration range of 0.25 to 25 ng/L, extending the low-end sensitivity substantially relative to published dilute-and shoot methods.<sup>1,2&nbsp;</sup></p><p>This performance enhancement simplifies operations by removing labor‑intensive extraction steps, while maintaining sensitivity required for regulatory guidelines thereby supporting high‑throughput PFAS workflows suitable for routine drinking water analysis. The PFAS compounds analyzed were PFBS, HFPO-DA, PFHpA, PFHxA, PFHxS, PFOA, PFOS, PFNA, and PFDA. These compounds were selected due to their relevance to ongoing regulatory actions and proposed maximum contaminant level (MCL) considerations for drinking water.&nbsp;</p><p>Specific highlights of this experiment include:&nbsp;</p><ul><li>Minimized peak tailing and solvent effects with 200 µL injections water/methanol (1:1) injections.</li><li>Increased throughput by elimination of solid phase extraction (SPE).&nbsp;</li><li>Enhanced sensitivity of 6495D LC/TQ at low end (0.25&nbsp;ng/L) for the dilute-and-shoot method in drinking&nbsp;water.</li></ul><h4><strong>Experimental&nbsp;</strong></h4><h5><strong>Instrumentation&nbsp;</strong></h5><p><a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/library?mainauthorItems=10&amp;search=+1290+Infinity+II"><strong>Agilent 1290 Infinity II LC</strong></a> coupled to <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/library?mainauthorItems=10&amp;search=6495D"><strong>Agilent 6495D LC/TQ system</strong></a> was utilized for the LC/MS/MS analysis. An Infinity&nbsp;II multisampler with an extended multidraw capillary tube (G7137-68307) was connected to the needle seat and injection valve with a union for large volume injection (200&nbsp;µL). The data were acquired with MassHunter acquisition software version 12.2. All LC/MS parameters are shown in Table 1 and 2. PFAS multiple reaction monitoring (MRM) transition methods were imported from the PFAS MRM database (G1736AA) while labelled homologues were optimized to obtain correct MRM transitions (Table 3).</p><h4><strong>Conclusion&nbsp;</strong></h4><p>This study demonstrates a practical large‑volume dilute-and-shoot workflow for the determination of PFAS in drinking water using the Agilent 1290 Infinity II LC coupled to the 6495D triple quadrupole LC/MS, and Agilent Altura Poroshell PFAS column. With 200 µL water samples diluted in methanol can be injected directly, eliminating the need for SPE concentration and significantly reducing sample preparation time. This method exhibits good sensitivity, accuracy, and reproducibility. The Altura PFAS column maintained consistent peak performance and retention stability even at low sub‑ppt concentrations, supporting long‑term reliability for trace‑level PFAS monitoring. Overall, the findings highlight the method potential to support routine, high‑sensitivity PFAS monitoring in drinking water laboratories.</p><h3><strong>2. KNAUER: Size matters - How to perform narrow calibration using the ClarityChrom® GPC Extension</strong></h3><ul><li>Technical note</li><li><a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/VTN_0049_knauer_size_matters_how_to_perform_narrow_calibration_using_the_claritychrom_gpc_extension_3cfdc7a45b.pdf"><strong>Full PDF for download</strong></a></li></ul><p>Gel Permeation Chromatography (GPC), also known as Size Exclusion Chromatography (SEC), is a separation technique that separates molecules based on their hydrodynamic size rather than chemical interactions. In a GPC column, separation is based on the ability of molecules to access the pores of the stationary phase. Smaller molecules can penetrate these pores and therefore spend more time inside the column, resulting in a delayed elution. Larger molecules, on the other hand, cannot enter the pores and pass through the column more quickly, leading to earlier elution.&nbsp;</p><p>As a result, molecules are separated by size in solution, providing the basis for the determination of molecular weight and molecular weight distributions. Calibration is therefore a fundamental requirement for ensuring the accuracy and reliability of the results. The ClarityChrom® GPC Extension enables users to efficiently create and apply different calibration models for precise analysis. Because D-amino acids are only present in trace amounts in fermented foods and biological systems, unlike L-amino acids, it is necessary to determine the concentration of each D-amino acid by separating them from the high concentrations of Lamino acids.&nbsp;</p><p>A comprehensive method for quantitating the thirty-seven D/Lamino acids was previously reported1), in which two chiral derivatizing reagents were employed to provide different diastereomers for each amino acid, with the instrument automatically switching between two separation methods to analyze each sample twice.&nbsp;</p><p>This article introduces an improved method that employs a single chiral derivatizing reagent to enable simultaneous analysis of D/L-amino acidsin a shorter time.</p><h4><strong>D/L-Amino Acid Analysis by UHPLC&nbsp;</strong></h4><p>In general, LC/MS or multi-dimensional LC is used to analyze D/L-amino acids by HPLC, because it is difficult to separate proteinogenic D/L-amino acids with a single separation mode. However, it is known that LC/MS is susceptible to matrix effects and less quantitative than HPLC. It is also known that the multidimensional LC method requires a long analysis time and very complicated HPLC setup. Therefore, a simple operational method that provides good separation for D/L-amino acids in a short time is needed.&nbsp;</p><p>For food analysis, a small particle column is used to achieve appropriate separation of the small amounts of D-amino acids from the large amounts of L-amino acids and co-existing contaminants. A <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/library?mainauthorItems=1&amp;search=X3"><strong>UHPLC system (Nexera X3</strong></a>) was employed in this study due to the increased system pressure caused by the use of a small particle column.</p><h4><strong>Conclusion&nbsp;</strong></h4><p>A method was developed for simultaneous separation of OPA/NIBC diastereomers for thirty-seven D/L-amino acids using a simple UHPLC system in approximately half the time of the conventional methods. By optimizing the derivatization reaction, all compounds were determined sensitively, accurately, and precisely. Separation of the target compounds from the contaminants was good in the liquor samples when evaluated using real liquor samples. It is expected to be easily applicable to D/L-amino acid profiling in food and beverage samples without using expensive MS detection or complicated multidimensional HPLC configurations as long as samples are prepared properly. Moreover, D/L-amino acid profiling could be used to research and develop foods containing D-amino acids intended to help to maintain and improve the intestinal environment, and could help in the early detection of diseases and determining optimal treatment strategies through biomarker discovery</p><h3><strong>4. PFAS are widely used in textiles to provide water, oil, and stain resistance, but increasing concerns about their environmental persistence and potential health effects have led to stricter regulations worldwide. California’s AB 1817 legislation, for example, limits TOF concentrations in textiles to 100 ppm beginning in 2025 and 50 ppm by 2027, creating a need for reliable analytical methods capable of assessing PFAS content in textile products.</p><p>The study evaluates a new <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/2150"><strong>Thermo Scientific Cindion Combustion Ion Chromatography System</strong></a>, which combines a Dionex Inuvion IC system with a Cindion combustion/absorption module. The system features an optimized Z-fold combustion tube design that improves combustion efficiency, shortens analysis time, reduces instrument footprint, and allows complete control through Chromeleon software. Unlike surface-sensitive techniques such as PIGE, the combustion-IC approach analyzes the entire sample, providing results that are independent of sample thickness. The method measures total fluorine (TF) after combustion and total inorganic fluorine (TIF) by direct IC analysis, with TOF calculated as the difference between TF and TIF.</p><p>To validate the approach, six textile samples representing waterproof and stain-resistant consumer products were analyzed and compared with results previously obtained using a Nittoseiko combustion-IC system. TOF concentrations ranged from approximately 1 ppm in one nylon fabric to nearly 375 ppm in a polyester baby bib. The values generated by the new Cindion system showed excellent agreement with the reference system, achieving between 100% and 105% of the previously reported results. Reproducibility was also strong, with relative standard deviations below 8% for all samples.</p><p>The authors conclude that the new Cindion C-IC platform provides an accurate, reliable, and efficient solution for PFAS screening in textiles. In addition to matching the performance of the established combustion-IC method, the enhanced system offers a flexible “2-in-1” configuration that allows seamless switching between combustion-IC and standalone IC operation. This improves workflow efficiency, reduces manual intervention, and provides manufacturers and testing laboratories with a practical tool for monitoring compliance with evolving PFAS regulations in textile products.</p><h3><strong>5. PFAS have been used in cosmetics to provide water resistance, oil repellency, or application smoothness. The unintentionally presented PFAS residues in raw materials may remain in finished cosmetic products, and can be absorbed through the skin. Lip products and spray type products can also lead to inhalation or oral exposure to residues.<sup>1,2</sup>&nbsp;</p><p>Agilent Captiva EMR PFAS Food cartridges are designed to provide streamlined and comprehensive matrix removal after QuEChERS extraction for complex sample matrices, demonstrating effective matrix cleanup efficiency and excellent performance for quantitative PFAS analysis.<sup>3,4</sup> In this study, we performed QuEChERS extraction followed by cleanup with Captiva EMR PFAS Food II cartridges for PFAS analysis in five types of cosmetics.</p><h4><strong>Experimental</strong></h4><h5><strong>Equipment and materials&nbsp;</strong></h5><p>The study was performed using an <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/library?mainauthorItems=10&amp;search=1290+Infinity+III"><strong>Agilent 1290 Infinity III LC</strong></a> system consisting of an Agilent 1290 Infinity III high-speed pump (G7120A), an Agilent 1290 Infinity III multisampler (G7167B), and an Agilent 1290 Infinity III multicolumn thermostat (G7116B). The LC system was coupled to an <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/library?mainauthorItems=10&amp;search=6495D"><strong>Agilent 6495D triple quadrupole LC/MS system</strong></a> (G6495D) equipped with an Agilent Jet Stream Electrospray ion source. Agilent MassHunter Workstation software was used for data acquisition and analysis.&nbsp;</p><p>The 1290 Infinity III LC system was modified using an Agilent InfinityLab PFAS Analysis HPLC conversion kit (part number 5004-0006), including an Agilent InfinityLab PFC delay column, 4.6 × 30 mm (part number 5062-8100). Chromatographic separation was performed using an Agilent <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/library?mainauthorItems=10&amp;search=ZORBAX+RRHD+Eclipse+Plus"><strong>ZORBAX RRHD Eclipse Plus</strong></a> C18, 95 Å, 2.1 × 100 mm, 1.8 µm (part number 959758-902).</p><h4><strong>Results and discussion&nbsp;</strong></h4><h5><strong>Chromatography and calibration curve linearity&nbsp;</strong></h5><p>Figure 2 shows the overlaid chromatograms for 41 PFAS calibration standards, including 0.05, 0.1, 0.2, 0.5, 1.0, 2.0, 5.0, and 10.0 ng/mL in ACN. The table on the right shows the peaks identification and corresponding retention time. Across 41 PFAS compounds, R² values ranged from 0.9940 to 0.9998, indicating excellent linearity for all analytes' calibration curves across the calibration range of 0.05 to 10.0 ng/mL.</p><h5><strong>PFAS detection in real cosmetic samples&nbsp;</strong></h5><p>Of the 15 cosmetic products assessed, one eyeshadow sample was detected with a trace level of positive PFAS analytes, including PFHeA, PFHpA, PFNA, and PFOA, at about 0.1 ng/g in the sample. Figure 5 shows the method blanks and positive detection of PFHpA and PFNA in this eyeshadow&nbsp;product.</p><h4><strong>Conclusion&nbsp;</strong></h4><p>A simplified, rapid, and reliable method using QuEChERS extraction followed by EMR mixed-mode passthrough cleanup using the Agilent Captiva EMR PFAS Food II cartridge and LC/MS/MS detection was developed for 41 PFAS analysis in cosmetics.</p><h3><strong>2. Metrohm: brightRC – Advanced brightener analysis with a response curve</strong></h3><p><strong>Next-level brightener determination in nonlinear systems</strong></p><ul><li>Application note</li><li><a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/an_v_242_download_83376b2923.pdf"><strong>Full PDF for download</strong></a></li></ul><h4><strong>WHAT IS BRIGHTRC?&nbsp;</strong></h4><p>brightRC is a response curve calibration approach for the determination of brightener-type additives by CVS or CPVS (Cyclic Pulse Voltammetric Stripping), hence the name «brightRC». This technique is suitable to measure brightener content in copper baths for plating. The method is based on signal normalization using the electrolyte value, which is the response for the virgin makeup solution (VMS) saturated with suppressor, and brightener defined as Q0 in the viva software. All recorded signals are divided by Q0 and the normalized values are plotted against concentration to generate an external calibration curve. This calibration can then be used for routine sample determination without repeated standard additions.&nbsp;</p><p>The sample is simply measured using the same method as the calibration, signals are normalized and evaluated using the mathematically fitted calibration curve. By decoupling calibration from sample measurement, brightRC removes the need for repeated standard additions with each sample measurement, enabling faster and more efficient routine analysis.&nbsp;</p><p>The brightRC calibration approach supports flexible regression models including quadratic and nonlinear regression. This allows reliable quantification of additive systems that do not exhibit a wide linear working range or exhibit nonlinear response characteristics, thereby expanding the range of applicable brightener chemistries.&nbsp;</p><p>One important detail that further differentiates brightRC from the MLAT approach relates to normalization of the signal for calibration and sample analysis. This normalization approach improves measurement robustness as variations in absolute signal intensity naturally occurring over the working day are mathematically compensated.</p><h4><strong>HOW DOES BRIGHTRC WORK IN PRACTICE?&nbsp;</strong></h4><p>The brightRC workflow separates calibration and sample analysis. It can be implemented in different setups (from manual to fully automated) depending on the additive system and concentration range. After recording the electrolyte value, calibration standards are measured to establish the calibration curve, otherwise known as the response curve. Once the calibration is stored, samples are determined with the same CVS or CPVS method, without further standard additions. The sample concentration is automatically calculated from the normalized signal using the calibration curve.&nbsp;</p><p>For sample measurement, viva 4.0 provides two practical approaches:&nbsp;</p><ol><li>«brightRC with solution exchange»&nbsp;</li><li>«brightRC with sample dilution» Both variants follow the same calibration principle and differ only in the sample handling strategy.</li></ol><h4><strong>WHEN TO USE BRIGHTRC?&nbsp;</strong></h4><p>brightRC is ideal for routine brightener control in stable plating baths where high sample throughput is required. It is particularly suitable when matrix conditions are consistent and electrode fouling is limited. The approach is especially advantageous for additive systems exhibiting nonlinear signal behavior. If strong matrix variations, significant breakdown products, or severe electrode fouling are expected and a linear correlation between signal and analyte concentration is guaranteed, standard addition techniques such as MLAT are preferred because they may provide additional robustness.</p><h3><strong>3. Shimadzu / ASMS: PFAS Analysis of Leachate: Evaluating Key Sample Preparation and LC–MS/MS Parameters for Improved Performance</strong></h3><ul><li>Poster</li><li><a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/WP_255_PFAS_Analysis_of_Leachate_Evaluating_Key_Sample_Preparation_and_LC_MSMS_Parameters_for_Improved_Performance_1114ee264c.pdf"><strong>Full PDF for download</strong></a></li></ul><p>Landfill leachate is a highly complex and variable matrix that poses significant challenges for the analysis of per- and polyfluoroalkyl substances (PFAS). Although U.S. Environmental Protection Agency Method 1633A provides a standardized framework for PFAS quantitation,1 analytical performance remains strongly influenced by sample preparation, chromatographic separation, and mass spectrometric conditions. Landfill leachate samples were extracted using WAX SPE following EPA 1633A and analyzed by LC–MS/MS using a <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/library?mainauthorItems=1&amp;search=LCMS-8060"><strong>Shimadzu LCMS-8060</strong></a><strong> </strong>under negative ESI conditions. This study evaluates the key factors governing LC–MS/MS sensitivity, signal response, and reproducibility in landfill leachate, including a systematic assessment of SPE, chromatographic behavior, and ionsource conditions across PFAS classes (Figure 1).</p><h4><strong>Results&nbsp;</strong></h4><p>Chromatographic performance was evaluated across four C18 columns (A–D) using peak area, height, asymmetry, and HETP. Two columns produced a higher signal and improved peak shape, while the remaining columns showed greater efficiency. Under repeated batches followed by storage in acetonitrile, the Shimadzu GIST column produced a consistent signal (≤15% change), whereas another column showed 20–50% signal loss, particularly for long-chain PFAS and precursors, indicating greater susceptibility to matrix accumulation (Figure 2).</p><h4><strong>Conclusion&nbsp;</strong></h4><ul><li>PFAS analytical performance was dependent on compoundclass-dependent extraction, chromatography, and ionization behavior.&nbsp;</li><li>GIST demonstrated lower signal variability under repeated matrix exposure, with lower signal degradation than alternative C18 phases.</li><li>Neutral PFAS showed poor retention on WAX sorbents, requiring optimization beyond standard EPA 1633A extraction conditions.&nbsp;</li><li>PFCA response was more influenced by gas flow conditions, whereas PFSA response was more sensitive to source temperature conditions.</li></ul><h3><strong>4. While established LC–MS platforms routinely support several hundred samples per day, further increases in throughput are often limited by chromatographic stability, robustness, and reproducibility under ultra-short gradient conditions, as well as the acquisition speed of the mass spectrometer. Here, we describe an advanced ultra-high-throughput LC–MS methodology developed on the Thermo Scientific Vanquish Neo UHPLC system and Thermo Scientific Orbitrap Astral Zoom mass spectrometer. The goal of this work was to extend the practical throughput limits of LC–MS while achieving deep proteome coverage and maintaining run-to-run reproducibility.</p><h4><strong>Materials and methods</strong></h4><h5><strong>LC-MS analysis&nbsp;</strong></h5><p>Samples were separated on a Thermo Scientific 150 µm × 70 mm C18 column (prototype, not commercially available yet) operated at 4 µL/min using a <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/398"><strong>Vanquish Neo UHPLC system</strong></a> in NanoCap Trap&amp;Elute configuration at room temperature. Prototype methods allowing for reduced LC duty cycles (approx. 1 min of sample pickup/loading and integrated trap-column offline wash) were applied, the standard gradients were adjusted to 1 – 3 min. The Thermo Scientific PepMap Neo 5 µm C18 300 µm × 5 mm cartridge was used as trap column and the separation column connected with a 15 µm ID capillary Thermo Scientific EASY-Spray emitter (ES994) to an EASY-Spray source. The <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/1959"><strong>Orbitrap Astral Zoom MS</strong></a> (ICSW AST 2.2) was operated in a dataindependent acquisition (DIA) mode, MS method parameters are given in Table 1. Comparative data for the chemoproteomics samples at lower throughputs (60-300 SPD) were acquired using an EASY-Spray 150 µm × 150 mm C18 column (ES906), as described before.&nbsp;</p><h5><strong>Data analysis&nbsp;</strong></h5><p>The raw data files were processed with Spectronaut® 20.5 software (Biognosys AG), <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/1782"><strong>Thermo Scientific Proteome Discoverer </strong></a>3.3 SP1 with <strong>CHIMERYS 5.0 </strong>or DIA-NN Enterprise 2.3.2 (Aptila Biotech GmbH) nodes using default settings as specified in the individual figures. The data was searched against the Homo sapiens SwissProt database (TaxID=9606: 42,252 sequences) and contaminants FASTA file. Peptide numbers correspond to unique stripped peptides</p><h4><strong>Conclusions&nbsp;</strong></h4><ul><li>The advanced Vanquish Neo UHPLC methodology in combination with the Orbitrap Astral Zoom MS delivered highly reproducible, deep and precise proteome profiling across ultra-high-throughput regimes (600 SPD) exceeding current field benchmarks.&nbsp;</li><li>The workflow enables rapid chemoproteomic screening, as demonstrated with activity-based protein profiling (ABPP) samples, reliably identifying enriched putative target proteins of an antibiotic small molecule probe.</li></ul><h3><strong>5. The study focuses on environmental exposure monitoring in Ghanaian firefighters and e-waste handlers, using ion mobility mass spectrometry to add separation power and improve identification confidence in complex biological matrices. The work is motivated by increasing regulatory concern around PFAS toxicity, bioaccumulation, and the need for reliable exposure assessment in human biofluids.</p><p>The experimental workflow combined reversed-phase LC with a quadrupole-cyclic ion mobility-time-of-flight MS system (<a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/202"><strong>SELECT SERIES</strong> <strong>Cyclic IMS</strong></a>). Human serum samples were prepared using SPE µElution plates with a polymeric reversed-phase/weak anion exchange sorbent. The LC system was modified with PFAS-free components and an isolator column to reduce background contamination, which is especially important for trace-level PFAS analysis. Identification was supported by retention time, accurate mass, isotopically labelled internal standards, and collision cross section values.</p><p>The results showed detectable levels of the four regulated PFAS in both occupational groups, with a combined detection rate of 83% across human serum samples and NIST reference standards. Firefighter samples generally showed higher concentration ranges than e-waste handler samples, especially for PFOS, PFOA, PFNA, and PFHxS. For example, PFNA reached up to 5.713 ng/mL in firefighter serum, compared with up to 1.513 ng/mL in e-waste handler serum.</p><p>Overall, the poster concludes that LC-Cyclic-IMS-MS provides a highly specific strategy for PFAS analysis in complex biological samples. Ion mobility and CCS values improve confidence in trace-level identification and help distinguish target PFAS from co-eluting matrix background. The authors suggest that this approach can support both targeted PFAS monitoring and broader non-targeted screening of known and emerging PFAS, while also helping future studies connect exposure data with environmental fate and potential toxicological effects.</p></div></div></div></div></div></div></section></div></div>
