Masaryk University

Masaryk University, based in Brno, is the Czech Republic's second-largest university. It has nine faculties and offers accredited study programs. Notably, it operates a polar station in Antarctica.

<p><strong>My name is James Little and my private consulting company is Mass Spec Interpretation Services. I contract for NIST, Wiley, and Sapec. Previously, I was a Research Fellow specializing in mass spectrometry at Eastman Chemical Company in Kingsport, TN and retired in early 2016 after &gt;37 years of service. For 2017-2019, I did NMR as a limited service employee specializing in end group determinations for Eastman.</strong></p>
<p>My main interest is the <strong>identification of organic compounds by mass spectrometry</strong> in organic mixtures.  All the technical information on my web page was approved as technical session <strong>handouts</strong>, conference <strong>presentations</strong>, <strong>workshops</strong>, <strong>journal publications</strong>, <strong>courses</strong>, etc. for distribution outside Eastman.  Other postings on this site are my own and do not necessarily represent Eastman’s positions, strategies or opinions.   I also include a “little” information about my sailing experiences.</p>
<p>In 2017, I purchased a used Model S85 Tesla and starting to participate in electric vehicle events.  In addition,  I am a member of the choir at St. Paul’s Episcopal Church, play competitive duplicate bridge, and race sailboats on Watauga Lake.</p>

<p><strong>I am a graduate of the University of Chemistry and Technology in Pardubice in the department of analytical chemistry. I have been involved in chromatography since the beginning of my professional career. For the last 26 years, I was working for Waters Corporation in the service department until my departure in 2023. As a service technician, I gained experience in laboratories all over the Czech Republic. Thanks to many years of experience, I can also offer knowledge from older instrumentation, whose official support has already ended. In addition to basic services, I can provide maintenance of chromatography systems, their confirmation and qualification.</strong></p>
<h3><strong>Service support of Waters HPLC a LCMS</strong></h3>
<h4><strong>1. On-site Service of HPLC and LC/MS</strong></h4>
<p>Performed the <strong>diagnostics</strong>, <strong>repair</strong> and <strong>testing after repair</strong>.</p>
<h4><strong>2. Performance Maintenance of HPLC and LC/MS</strong></h4>
<p>The condition evaluation of the chromatography system by performing targeted tests. Proposal for <strong>partial or complete maintenance</strong> according to the manufacturer&#39;s recommendations. Performed tests after maintenance.</p>
<h4><strong>3. HPLC and LC/MS systems confirmation, calibration</strong></h4>
<p>At the beginning is basic testing of the individual modules of the chromatography system. After that, if necessary or according to request, the maintenance is performed. <strong>The required parameters are calibrated</strong>. This is followed by <strong>tests verifying the basic characteristics</strong> of the accuracy and reproducibility of the system and is evalueted as to whether they meet the specification. Certified chemicals and calibrated tools are used to confirm the system.</p>
<h4><strong>4. Installation and commissioning of older HPLC and LC/MS systems</strong></h4>
<p>In the case of <strong>older used chromatographic instruments</strong>, their condition is evaluated. If necessary, then their repair, maintenance, or addition of missing components will be proposed. Confirmatory tests including  accuracy and reproducibility evaluation characteristics are used for verification. After <strong>complete installation</strong> and <strong>documentation of test results</strong>, operator <strong>training is carried out</strong>. This can be followed by the <strong>preparation of the system for targeted application</strong> and, in cooperation with application support, <strong>the development and optimization of the chromatographic method</strong>.</p>
<h4><strong>5. HPLC and LC/MS systems qualification</strong></h4>
<p>To successfully complete all qualification tests, the system must be in good condition. Therefore, at the beginning, its condition is verified by evaluating its condition since the last maintenance. <strong>The maintenance and other tests are performed</strong>, such as <strong>pressure tests</strong>, checking <strong>the sensitivity of the detectors</strong>, <strong>MS charging</strong> test, etc. If necessary or according to a previous order, repair/maintenance is performed. During <strong>the qualification process, all the necessary parameters are verified</strong>, which are then evaluated.</p>
<p>Both the parameters of <strong>individual modules (OQ)</strong> and <strong>the entire system are qualified</strong>. Certified chemicals and calibrated tools are used for the system qualification. The customer supplies only the mobile phase.</p>
<h4><strong>6. Instruments and software operation training of Waters HPLC and LCMS</strong></h4>
<p>The training is provided according to the <strong>customer&#39;s needs and request</strong>.</p>

28th International Symposium on Separation Sciences: ISSS 2024

<h4><strong>Overview</strong></h4>
<p>The International Symposium on Separation Sciences (ISSS) is a prominent annual conference that brings together researchers, practitioners, and experts from various fields involved in separation sciences. <p>The International Symposium on Separation Sciences (ISSS) series started in 1990 in Bled (Slovenia) as the continuation of a series of local seminars and conferences held in Zagreb since 1967. The first six editions were alternatively organized in Slovenia and Croatia and the subsequent editions were held as annual international meetings in Central European Countries, under the auspicious of the <a href="https://cegss.ptchem.pl/"><strong>Central European Group for Separation Sciences (CEGSS)</strong></a>. The symposium traditionally offers an opportunity to scientist and practitioners of high-performance separation techniques, such as GS, HPLC and capillary electromigration techniques, to present and discuss the latest progresses related to all aspects of these separation techniques, their hyphenation to other separation methods or spectroscopic techniques, and their applications in scientific and industrial areas.</p>

<p><strong>The Institute of Organic Chemistry and Biochemistry of the Czech Academy of Sciences (IOCB Prague) is a leading scientific institution in the Czech Republic, recognized internationally. Its primary mission is basic research in the fields of chemical biology and medicinal chemistry, organic and material oriented chemistry, chemistry of natural compounds, biochemistry and molecular biology, physical chemistry, theoretical chemistry, and analytical chemistry.</strong> </p>
<p>Today, over 900 employees including 220 PhD students in 49 research and service groups are involved in a broad spectrum of disciplines, which results in more than 250 publications per year, including papers in the most cited prestigious journals such as Nature or Science.</p>
<p>The Institute’s emphasis on interaction between chemistry and other sciences leads frequently to medicinal applications. An integral part of the mission of the Institute is, therefore, to transfer the scientific results into assets which help people to live better lives. </p>

<h3><strong>Highlights</strong></h3>
<p><strong>Successful basic research and modern structure</strong></p>
<p>The quality of IOCB science is regularly and rigorously assessed by an international advisory board comprised of respected authorities from research institutes and universities around the world. IOCB has pioneered a progressive system of junior groups with talented scientists from different countries hired in an open international competition, during which they demonstrate their unique expertise and submit scientific proposals. Every five years, all research groups undergo a complex evaluation that strictly adheres to transparent rules based on scientific merit.</p>
<p><strong>Medicinal chemistry and drug development</strong></p>
<p>IOCB has always been active and successful in applied research and practical applications, particularly in medicinal chemistry. The tradition started in 1969 with an ointment called Dermazulen, which was followed by the development of several human peptide hormones and their analogues (lysin-vasopressin, oxytocin).</p>
<p>From a global perspective, the most significant contributions were acyclic nucleotide phosphonate antivirals (especially tenofovir as a component of Truvada, Atripla, and other anti-HIV and anti-HBV drugs) discovered by Prof. Antonín Holý at IOCB and later developed and marketed by Gilead Sciences, Inc.</p>
<p>Besides this well-known antiviral drug story, several other nucleoside compounds developed at IOCB became approved drugs. These include Decitabine, which is used in the treatment of acute myeloid leukemia, Azacytidine, which targets myelodysplastic syndrome (both discovered by Holý’s peer Dr. Alois Pískala), and 9-(2,3-dihydroxypropyl)adenine (DHPA), an acyclic nucleoside analogue discovered by Holý and used clinically in anti-herpes ointments.</p>
<p><strong>Gilead Sciences Research Centre at IOCB Prague</strong></p>

<p>The unique collaboration between IOCB Prague and Gilead Sciences, Inc. dates back to the 1990s. A partnership program called Gilead Sciences Research Centre at IOCB Prague was established in 2006 for an initial period of five years with an annual donation of $1.1 million to expand IOCB efforts in the field of human disease research. The program was renewed in 2011 and again in 2016 with increased annual funding of $1.35 million.</p>
