What exactly is a “source” in mass spectrometry?</strong></h3><p>Whether you’re running LC-MS, imaging-MS, or native-MS, the ion source is where molecules leave the chromatograph, capillary, or sample stage and become charged gas-phase particles that the mass spectrometer can detect. Why electrospray changed the game</strong></h3><p>In 1988 John B. Fenn demonstrated that a fine, high-voltage spray could coax intact proteins into the gas phase (see figure). The work earned a share of the 2002 Nobel Prize in Chemistry. Because electrospray produces multiply charged ions, large biomolecules fit within the m/z window of modern analyzers. Electrospray 101: What to Know About LC-MS Sources – and a First Look at Optispray

<h4><strong>Traceability in the 19th century&nbsp;</strong></h4><p>Once a dairymaid bought one kilo of flour from the local shop. Next day she returned to sell one kilo of butter to the shopkeeper. He then complained that 50 grams of the kilo were missing.&nbsp;</p><blockquote><p>Oh that is odd, the dairymaid said, to get the correct weight I used the kilo of flour you sold to me yesterday to weigh the butter!&nbsp;</p></blockquote><h4><strong>Traceability today&nbsp;</strong></h4><p>Traceability is the ability to trace, for example, the ancestors of a person or the origin of a product. The word can be derived from the Latin tractus = drawn or the verb trahere = to draw.&nbsp;</p><p>Traceability can refer to documentation (e.g. a sampling procedure), a laboratory, analyst or method. But as in ISO/IEC 17025, this leaflet deals with traceability of measurement results. That is, measurement results have to be traceable to the metrological<sup>1</sup> references <strong>used</strong>. To be specific, we use the term <i><strong>metrological traceability</strong></i>.&nbsp;</p><blockquote><p>The current VIM<sup>2</sup> definition of metrological <strong>traceability is: </strong>property of a measurement result whereby the result can be related to a <strong>reference</strong> through a documented unbroken <strong>chain of calibrations</strong> each contributing to the measurement uncertainty<sup>2</sup>&nbsp;</p></blockquote><h4><strong>The references&nbsp;</strong></h4><p>Ideally the <strong>references</strong> should be the values of national and international standards expressed in SI units. The traceability is realised through chains of <strong>calibrations</strong>, the traceability chain. The result is reported as total Hg on dry weight basis (105 ºC, 12 h determined on a separate sample portion) and the measurement uncertainty is reported with a 95 % level of confidence (coverage factor, k=2). In this case mercury was determined with a mercury analyser (cold vapour atomic spectroscopy) after a microwave digestion.&nbsp;</p><p>The samples are weighed on a balance with a calibration certificate relating the weight measured to the SI unit of mass (kg).&nbsp;</p><p>The acid digest is diluted in a volumetric flask where the manufacturer documents the traceability of the volume of the flask to a national standard.&nbsp;</p><p><i><strong>The calibration curve was prepared using a CRM (certified reference material) – a mercury standard with a certificate stating a quantity value of 0.998 ± 0.005 mg/L (k=2) and with traceability to pure mercury.&nbsp;</strong></i></p><p>The method is validated using an appropriate matrix CRM with a total mercury mass fraction of 1.97 ± 0.04 mg/kg (<i>k</i>=2). The purpose of the validation is to check the method performance.</p><p><img src="https://storage.googleapis.com/labrulez-bucket-strapi-h3hsga3/Eurachem_Metrological_Traceability_of_Analytical_Results_4_1ef2bc4b97.jpg" alt="Douglas Hasbun: An “analyst” knowing the traceability of his/her values for volume, time, mass, calibration solution concentration and temperature" srcset="https://storage.googleapis.com/labrulez-bucket-strapi-h3hsga3/thumbnail_Eurachem_Metrological_Traceability_of_Analytical_Results_4_1ef2bc4b97.jpg 119w," sizes="100vw" width="808" height="1062"></p><h4><strong>How can a laboratory demonstrate metrological traceability?&nbsp;</strong></h4><p>The evidence required by the laboratory to demonstrate traceability for the mercury result is shown below:&nbsp;</p><ol><li>concentration of the Hg solution – a certificate for the CRM solution;&nbsp;</li><li>mass of sample – calibration certificate for the balance;&nbsp;</li><li>volume of volumetric flask – calibration certificate of the manufacturer;&nbsp;</li><li>drying temperature – calibration of oven temperature;&nbsp;</li><li>digestion conditions – calibration of e.g. temperature;&nbsp;</li><li>drying time – ordinary clock or stopwatch&nbsp;</li></ol><p>Point 1 would need special attention to assure the quality and traceability for the calibration standard. Traceability for points 2, 3 and 6 is easily achieved with adequate uncertainty using commercial equipment.&nbsp;</p><p>Points 4 and 5 need additional attention by the lab.&nbsp;</p><p>The use of the matrix CRM in the method validation is vital but not part of the traceability chain, since it is not used for calibration. If this CRM is used for recovery correction it should be part of the traceability chain.&nbsp;</p><h4><strong>Measurement quality&nbsp;</strong></h4><ul><li>Method <strong>validation</strong> demonstrates that the method (set of conditions) used in a particular laboratory at a certain time was fit for purpose and all significant effects were identified.&nbsp;</li><li>Calibration of critical equipment completes the metrological<strong> traceability </strong>chains.</li><li><strong>Measurement uncertainty </strong>is estimated from the method validation and the traceability.&nbsp;</li><li><strong>Quality control </strong>(internal and external) assures that the measurement results (including uncertainty) are of the same quality as at the time of validation.</li></ul><h4><strong>Conclusion</strong></h4><p>The <strong>traceability of a measurement result </strong>refers to <strong>metrological traceability</strong> as defined by VIM. It relates the result to SI units or other agreed standards/references.&nbsp;</p><p>Traceability is essential for comparability of results and is a requirement of ISO/IEC 17025. Traceability is easily achieved following good laboratory practice.</p>

<p>David will be your host for this event. He completed his PhD in Biochemistry at Nottingham Trent University, focused on the effects of toxins on glioma cells. After this he joined Lonza to work as a research and development scientist using different analytical techniques to characterize antibodies and proteins. Later he worked for Thermo Fisher Scientific as applications specialist and Biopharma Expert, and as business development manager in two CDMOs,  focusing on antibodies and viral vector projects. David joined Waters in June 2022 as principal biopharma market development manager.   </p>

<p>Emma completed her PhD in Analytical Chemistry at Loughborough University, focused on the development of ion mobility mass spectrometry for complex mixtures.</p>
<p>After her post doctoral research at University of Leicester and having worked as analytical scientist at Hall Analytical, using a variety of analytical techniques, Emma joined Waters in June 2019 as a senior applications chemist in the Demo lab in Wilmslow. In 2021 she was promoted to European applications lab manager.</p>

<p>Chris is currently working as a Principal Domain Specialist (Applications science) at Waters, having been with the company for 5 years.  He works with the Waters informatics teams to help deliver the waters_connect software applications. His focus has been on the CONFIRM Sequence application.  His background is in Bioprocessing, having worked at FUJIFILM Diasynth Biotechnologies for 3 years as a principal scientist in Mammalian cell culture, following completion of a PhD in mammalian cell line engineering at the University of Aberdeen in 2016.</p>
<h5><strong>Ali Tiss</strong> (Sales Specialist Biopharma Chemistry, Europe South, Waters Corporation)</h5>
<p>Ali completed his Ph.D. in biochemistry and structural biology at the Mediterranean University (CNRS, Marseille, France), where he contributed to the R&amp;D of the anti-obesity drug, Xenical®, developed by Roche Ltd. Then, he spent about 20 years of academic research (France, UK, Kuwait) in biochemistry, liquid chromatography, mass-spectrometry, and proteomics, focusing on detecting and characterizing biomarkers related to human diseases (cancer, diabetes, obesity). Ali joined Waters in October 2021 as a BioPharma and New Modalities Specialist in Chemistry for Southern Europe.</p>

<p><strong>This presentation will focus on our use of a RapidFire 400 High Throughput Mass Spectrometry system coupled to a triple quadrupole mass spectrometer, to build and implement a single platform for the rapid biochemical characterization of multiple biological enzyme targets.</strong></p>
<p>Because of the high sample throughput compared to traditional LC-MS/MS, we can comprehensively characterize the activity of multiple enzymes across a variety of substrates including biologically relevant substrates (e.g. oligonucleosomes) and tailor our Medicinal Chemistry small molecule compound design efforts to target the enzyme activity of interest.  Employing a single platform with internal redundancy, enables us to both quickly and confidently develop suites of assays within and across multiple enzyme classes, without many of the limitations present with other technologies.</p>

<h3><strong>Part 2: Analytical Innovations to Speed-up Antibody Characterization</strong></h3>
<p>The characterization biotherapeutics is essential to assess their safety, quality, and efficacy. In this presentation, we show novel technologies that can address the growing demand for comprehensive and faster characterization of multi-specific antibodies. In the first example, Mobile Affinity Selection Chromatography is used to rapidly assess antibody titer and aggregation levels of crude culture filtrate. We take advantage of this method for unbiased analysis of mispaired chains and aggregates directly from culture supernatants obviating a protein A clean-up step. We show fast aggregate and titer molecular structure quantifications without sample preparation can be achieved with multi-sampler overlapped injections, dramatically improving analysis throughput (~ 2.5 min per sample) without carryover. In the second example, mAb digestion process (&gt; 30 min) is accelerated on the surface of microdroplets (&lt;1 ms) (Anal. Chem., 2021, 93, 3997-4005). We show automated microdroplet reactions under native-pH conditions by combining programmed flow injection with a JetStream ionization source. Intact subunit mass analysis and middle-down characterization of antibody subunits are completed every 2 min. (Anal. Chem., 2023, 95, 3340-3348). Finally, we propose droplet reactions every 2-4 seconds per antibody sample using RapidFire BLAZE mode for ultrafast sample introduction and digestion. We envision, that these analytical innovations will enable comprehensive and faster analysis of biotherapeutics.</p>

<p>Harsha Gunawardena obtained his Ph.D. in Analytical Chemistry from Purdue University and a Master of Science degree in Analytical Chemistry from University of North Carolina. He was a post-doctoral fellow at the Lineberger comprehensive cancer center, UNC school of medicine where his research focused on refining quantitative platforms for large-scale clinical proteomics and proteogenomic analysis. He was a co-investigator for the NCI clinical proteomics tumor analysis consortium (CPTAC), the ENCODE consortium. His industry career as scientist started at Bayer Health Care providing mass spectrometry and bioanalytical support of plasma-based therapeutics. He later worked at Amgen applying proteomics methods to support biomarker programs and discovery programs in oncology and inflammation. In his current role at Janssen R&amp;D, Johnson &amp; Johnson, he supports discovery early development programs that span several therapeutic modalities.</p>

<p>These new webinars will focus on the analytical separation of biomolecules such as peptides and amino acids.</p>
<p>Reverse-phase chromatography is the predominant technique used for the analyses and or purification of chemically synthesized peptides or peptides generated by the enzymatic digestion of proteins (i.e., peptide mapping). It is advantageous to have a simplified step-by-step approach to develop a method that can be applied to chemically synthesized peptides or protein digests.</p>
<p>Amino acid analysis is required in several different areas of research and is a fundamental tool in various product analytical activities.  Due to the varied properties of amino acids (i.e., acidic, basic, etc.), the separation and detection represents an analytical challenge and depending on throughput, sample preparation can be laborious.  </p>

UNDERSTANDING AMINO ACID ANALYSIS

<p><strong>On December 1, 2022 a harmonized standard for General Chapter 621 Chromatography was released. This standard incorporates the USP with 2.2.46. Chromatographic Separation Techniques European Pharmacopoeia (EuPh) and 2.01 Liquid Chromatography Japanese Pharmacopeia (JP) texts. Harmonization of these regulatory guidance provides increased method flexibility through the employment of modernized of chromatographic tools without the need for full monograph re-validation.</strong></p>
<p>The extent to which the various parameters of a compendial chromatographic test may be adjusted without fundamentally modifying the pharmacopeial analytical procedures is defined in U.S. Pharmacopeia (USP) General Chapter 621 Chromatography.</p>
<p>In this presentation, we combine the gradient method adjustments described in this chapter to achieve both column and system modernization for the USP monograph separation of antiviral drug abacavir sulfate. </p>
