<h4>Introduction&nbsp;</h4>Millions of tests, measurements and examinations are made every day in thousands of laboratories around the world. There are innumerable reasons underpinning them, for example: as a way of valuing goods for trade purposes; supporting healthcare and construction; checking the quality and safety of food and feed; and in forensic analysis and environmental monitoring. Virtually every aspect of society is supported in some way by analytical work.&nbsp;The cost of carrying out these measurements is high and additional costs may arise from decisions made on the basis of the results. For example, tests showing food to be unfit for consumption may result in compensation claims. In addition, tests confirming the presence of banned drugs could result in fines or imprisonment. Clearly it is important to make a correct measurement and be able to show that the result is correct.<h4>The validation/verification process</h4>Most analysts know that method validation/verification is important, but exactly why, how and when it should be done is not always clear.&nbsp;Method validation/verification is the process whereby the laboratory demonstrates whether or not a method is ‘fit for purpose’ (Fig. 1). This means that the tests carried out should be appropriate with respect to uncertainty, cost, time etc. The final report should present analytical data in such a way that the customer can, readily, interpret it and draw appropriate conclusions.<img src="https://storage.googleapis.com/labrulez-bucket-strapi-h3hsga3/Eurachem_The_validation_verification_82742c1029.jpg" alt="Eurachem: Fig. 1. The Guide and its supplements [1-4]:&nbsp;<ul><li>Introduce the basic concepts related to method validation and method verification;&nbsp;</li><li>Indicate how to plan, record and report validation and verification studies to best support the statement of ‘fitness for purpose’;&nbsp;</li><li>Provide key definitions and the rationale behind the experiments for assessing the various performance characteristics and related topics (Fig. 3);&nbsp;</li><li>Include quick reference tables that suggest experiments together with the necessary statistical calculations for evaluation and reporting each performance characteristic;&nbsp;</li><li>Provide support to the analyst on how to make the best use of method validation data for setting up an internal quality control plan.</li></ul><img src="https://storage.googleapis.com/labrulez-bucket-strapi-h3hsga3/Eurachem_The_Fitness_for_Purpose_of_Analytical_Methods_95fc1a74e3.jpg" alt="Eurachem: Fig. 2. Any method validation or verification study will require the laboratory to investigate several performance characteristics:<ul><li>Precision<ul><li>Repeatability&nbsp;</li><li>Intermediate precision</li></ul></li><li>Trueness</li><li>Selectivity</li><li>Working range</li><li>Analytical sensitivity</li><li>Limit of detection</li><li>Limit of quantification</li><li>Ruggedness</li></ul>Exactly which characteristics are studied will depend on the analytical application. Verification of the performance of a standard method requires substantially less work than validation of a method developed in-house. Legislative/sectoral requirements must also be considered.&nbsp;Sampling and subsampling can be part of the measurement or testing procedure and must, in those cases, be validated.

Sepsis is a growing global challenge due to factors such as high mortality rate and the increasing prevalence of antimicrobial resistance.
<h4>A single and rapid Zeno SWATH DIA proteomic analysis for concomitant identification and antibiotic resistance profiling of micro-organisms from positive blood cultures</h4>

Today, approximately 1.3 million people die each year worldwide from bacterial antimicrobial resistance (AMR). AMR is of great concern in the context of bloodstream infections (BSIs) when it evolves into sepsis. If MALDI-TOF has markedly shortened the delay of the pathogen identification step, AMR evaluation is still based on growth tests in the presence of antibiotics. With the aim of streamlining this turnaround time to less than 90min from a positive blood culture, the LC/Zeno SWATH DIA sensitivity and specificity performances were evaluated to concomitantly provide the identity and antibiotic resistance profile of etiological agents related to BSIs based on peptide surrogates.
<h4>Plasma proteomic landscape and patient heterogeneity in sepsis revealed by high-throughput mass spectrometry</h4>

Incomplete knowledge of the sepsis plasma proteome has limited our understanding of pathophysiology in the heterogeneous syndrome of sepsis. Here we apply high-throughput liquid chromatography – tandem mass spectrometry to delineate the plasma proteome for sepsis and comparator groups, involving 2622 samples in a single batch. We show how this scale of data can establish shared and specific proteins, pathways and co-expression modules in sepsis, and be integrated with paired leukocyte transcriptomic data. This work reveals novel subphenotypes informative for the response state and differential outcome, highlighting opportunities for a systems-based precision medicine approach to sepsis.

His activities from 1997 until 2021 had focused on providing method and technical application support for dissolution and drug release testing in the pharmaceutical industry as well as working with VanKel, Varian and eventually Agilent R&amp;D, Marketing and Sales teams. Prior to 1997, Bryan performed analytical testing, supervised, and managed pharmaceutical laboratories with Bristol-Myers Squibb, Merck, AAI and Abbott Laboratories until he joined the dissolution equipment manufacturer VanKel.

Bryan has developed and routinely conducts numerous seminars related to dissolution and drug release testing. He’s provided numerous talks at exhibitions and conferences and has published numerous articles in journals and book chapters on dissolution related techniques and topics. Bryan has been a former member of the USP Biopharmaceutics Expert Committee and is presently a member of several USP Expert Panels on Performance Verification Testing, Capsules, Performance Tests for Semisolid Dosage Forms and Solubility Criteria for Veterinary Drugs. He’s a Steering Committee member of the AAPS In-Vitro Release Dissolution Testing (IVRDT) Community Leadership Team, co-founder and co-moderator of the Dissolution Discussion Group (DDG) and is a current member of the American Chemical Society.
