While they may appear simple, their formulation is actually quite sophisticated. Each gummy is produced using a precisely balanced combination of gelling agents, sugars, acids, flavourings, and colourants, all working together to deliver the desired taste and texture.For instance, the characteristic chewiness of gummy sweets results from the interaction between gelatin and sugars. Even minor adjustments—such as changing the type or concentration of the gelling agent—can significantly alter the final product. In addition, manufacturers must meet regulatory and labeling requirements, including certifications like Halal compliance. This makes accurate and consistent monitoring of ingredients essential throughout production.<h4>The importance of SEC fingerprinting</h4>Size Exclusion Chromatography (SEC), also known as Gel Permeation Chromatography (GPC), offers a fast and reliable way to analyze gummy formulations. One sample contained porcine gelatin, while the other used bovine gelatin. The resulting SEC fingerprints showed clear differences in the gelatin region, allowing unambiguous identification. The use of alkylamine ion pair and perfluorinated alcohols in the analysis of oligonucleotides by LC-MS techniques has been the mainstay in characterisation and quantitation methods for synthetic oligonucleotides for decades. Often, analytical chemists default to relatively high concentrations of both, but how much ion pair is needed for optimal chromatographic performance and how much does perfluorinated alcohol concentration contribute to electrospray ionisation efficiency?
In this presentation, we provide an overview of nucleic acid therapeutics and discuss the chromatographic challenges associated with oligonucleotides. We also explore modulation of the mobile phase additives alkylamine ion pair and perfluorinated alcohols. Finally, we will show how optimisation of mobile phase can improve not only chromatographic performance and MS sensitivity, but may also directly impact MS data acquisition, thus facilitating data analysis.

The software includes MS Search, MS Interpreter, and AMDIS plus ChemSketch for structural drawing. The series should be useful to a wide variety of research and developments scientists involved in medical, environmental, industrial, forensic, and biological disciplines.
Initial sessions will primarily discuss electron ionization gas chromatography/mass spectrometry (EI GC/MS) data. However, many of the principles are applicable to tandem spectra obtained by liquid chromatography/mass spectrometry (LC/MS) which will be discussed in the latter part of the series.
