At the January 2025 CFSRE Seized Drugs Symposium, a Thermo Fisher Scientific–sponsored workshop showcased the potential of <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/2240">Thermo Scientific VeriSpray PaperSpray mass spectrometry </a>(PS-MS) to transform drug analysis workflows in both forensic and clinical toxicology. Stefania’s PhD research seeks to change that. Her team is developing a rapid, extraction-free method using VeriSpray PS-MS for the targeted analysis of plasma samples from patients presenting with acute drug toxicity.By using dried matrix spots on a paper substrate and coupling this with a <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/404">Thermo Scientific TSQ Altis Plus Triple Quadrupole Mass Spectrometer</a>, Stefania’s work provides an alternative screening technique to chromatography, delivering faster, actionable results to guide early clinical decision-making.<h5>Key takeaways:</h5><ul><li>Paper spray MS is an ambient ionization technique suitable for emergency toxicology due to its rapid results and minimal sample prep.</li><li>Enhancing the paper substrate and adding microextraction steps can boost sensitivity without sacrificing speed.</li></ul><h3>Substrate matters: boosting detection with paper treatment</h3>One key limitation of PS-MS is the interference that can arise from the paper substrate itself. Marwa’s research explored nine different paper treatments, ranging from acids to solvents, to improve the signal-to-blank (S/B) ratio when analyzing eight common drugs in whole blood.The most promising results came from formic acid and nitric acid treatments, which significantly enhanced analyte detection. Although these methods are currently time-consuming, Marwa envisions future industrial-scale production of pre-treated cartridges to support scalable clinical workflows.<h5>Key takeaways:</h5><ul><li>Substrate optimization can significantly reduce background noise in PS-MS analysis.</li><li>Acid-treated paper enhances S/B ratios, enabling more reliable detection of drugs in blood.</li><li>Industrial pre-treatment could make these improvements broadly accessible.</li></ul><h3>DLLME meets paper spray: improving sensitivity in plasma</h3>Saeed Almheiri’s presentation focused on dispersive liquid-liquid microextraction (DLLME) as a sample prep technique to increase sensitivity for PS-MS analysis of drugs in plasma, without adding significant complexity to the workflow.Using DLLME prior to VeriSpray analysis, Saeed demonstrated improved detection limits (as low as 0.3 ng/mL) across a panel of 10 drugs, including buprenorphine, cocaine, and MDMA. Importantly, this method maintained the fast, minimal-prep advantages that make paper spray ionization so appealing. She drives the implementation of ICH Q2(R2), Q14, and Analytical Quality by Design (AQbD) principles in collaboration with global biopharmaceutical partners. Amanda serves as a member of the United States Pharmacopeia (USP) Pharmaceutical Analytical Lifecycle and Data Science Expert Committee and is a board member of the ECA Analytical Quality Control Group. Before joining RIC, she spent more than 12 years at USP, where she worked both as a Senior Scientist – leading method development and validation projects, impurity analysis by LC-MS/GC-MS, and reference standards characterization by LC-HRMS – and as a Scientific Affairs Manager, contributing to major standards such as &lt;1220&gt; and &lt;1225&gt; and driving initiatives in AQbD, predictive modeling, and analytical procedure lifecycle management. In this series, you will learn the fundamentals of the classical applications of CE.

Per- and poly-fluoroalkyl substances (PFAS) are persistent chemicals commonly found in surface protectants and fire-fighting foams. Research has revealed that exposure to environmentally relevant concentrations of some PFAS can reduce reproductive success and have other adverse effects on animals. In this webinar, Professor Todd Anderson, from the Department of Environmental Toxicology, Texas Tech University, presents new findings from field studies determining PFAS concentrations in plant and animal tissues. His team have been involved in laboratory and field studies where it was necessary to determine PFAS concentrations in plant and animal tissues to (1) better understand the pathways of PFAS trophic transport from soil to higher organisms, and (2) to determine PFAS burdens in exposed organisms and the potential for maternal transfer of PFAS from adults to offspring.
LC-MS analytical methods for PFAS determination in water and soil matrices provide a solid foundation upon which to make slight changes to improve method performance for extracts of plant and animal tissues. In controlled laboratory studies with an avian model, we’ve used these modified methods with varying degrees of success, to measure PFAS in adult livers, eggs, hatchling livers, and brain tissue (adults and hatchings). These PFAS measurements in tissues allowed for determination of avian Toxicity Reference Values based on survival, growth, and reproduction endpoints, as well as maternal transfer from exposed adults to unexposed hatchlings. In field studies on historical fire training areas, we’ve used these modified methods on a limited set of PFAS analytes, to measure PFAS in vegetation, invertebrates, and small mammal livers. These PFAS measurements have allowed us to better understand the trophic transport pathway(s) from soil to higher organisms based on diet (herbivores, insectivores, omnivores) or activity (grooming, nest building).

High temperature gel permeation chromatography (GPC) system coupled with a viscometer enables the detailed analysis of polyolefins.

The PFAS global research and testing community are now being asked to fully characterize PFAS in samples. Targeted analysis relies on a limited number of available reference standards, however it is well known that thousands of PFAS compounds and transformation products exist in the environment. To support laboratories, Thermo Fisher Scientific has worked with collaborators to develop a novel workflow to provide a better understanding of the diversity of PFAS contamination in samples, and at the same time provide meaningful targeted quantitative results in a variety of matrices.
In this webinar, our experts will show a simultaneous targeted quantitative and non-targeted analysis method, with quantitative results down to part-per-trillion levels, for a 250 mL water sample prepared by EPA 1633 protocols. Additionally, non-targeted data is presented for food contact materials and diluted AFFF sample matrices to demonstrate the applicability of the method to solid samples.

Throughout his career he has published more than 20 scientific publications in the area of mass spectrometry, altogether having more than 1,000 citations.
