Tips for Developing Solid Phase Extraction Methods

<p><strong>Activated carbons are widely used for purification and separation applications. However, they are not employable as chromatography media due to the lack of chemical purity, irregular shape, poor mechanical integrity, and lack of reproducibility.</strong></p>
<p>Carboxen® synthetic carbon adsorbents are a synthetic form of activated carbon. These synthetic carbons are derivatized resins. Due to their high purity, spherical shape, and outstanding mechanical properties, Carboxen® synthetic carbon adsorbents are well suited for use in low-pressure liquid chromatography (LPLC) applications. This chromatography media can withstand pressures up to 110 MPa, which is several orders of magnitude greater than other LPLC media. The tapered pores improve the adsorbent’s kinetic and thermodynamic efficiency, enabling greater capacity while operating at increased throughputs. The highly engineered, derivatized resins bring value in both adsorption and hydrophobic interaction chromatography.</p>
<p>These media bring value to a wide range of LPLC applications for both small and large molecule separations. As example, these adsorbents are used for clearance of high-risk impurities in biochemical purifications such as host cell proteins, trace metals, and extractables and leachables. These synthetic carbons have been trusted by NASA on several missions including their use on the International Space Station.</p>

<p><strong>Given the differences in physicochemical properties and concentrations of metabolites in biological systems, getting a broad metabolite coverage in untargeted metabolomics is always challenging in terms of the instrumental techniques that are necessary.</strong></p>
<h5>Two approaches are currently in use:</h5>
<ol>
<li>Using LC/Q-TOF MS for lipidomics; GC/MS for small metabolites that can be made volatile by derivatization and CE/MS for polar and ionic compounds.</li>
<li>LC/Q-TOF MS for lipidomics and LC/QQQ MS for polar and semipolar compounds.</li>
</ol>
<p>Even a third approach with LC/Q-TOF MS using either C18 column for lipids or HILIC for more polar compounds could be used.</p>
<p>Advantages and limitations of these analytical platforms will be discussed with some examples from our laboratory.</p>

For his graduate work, he studied with Professor Peter Carr in the Chemistry Department at the University of Minnesota from 2001–2007, where he worked on the development of fast, comprehensive two-dimensional liquid chromatography.</p>

<p>In the fall of 2008, Stoll was hired as an Assistant Professor in the Chemistry Department at Gustavus Adolphus College, where he mainly teaches quantitative and instrumental analysis courses. In 2014, he was promoted to Associate Professor, and in 2018, he was promoted to Professor. Currently, he is co-chair of the Chemistry Department. Active research projects in his laboratory touch upon most aspects of multi-dimensional separation methodologies, including optimization strategies, characterization of selectivity in reversed-phase HPLC, instrument development, and applications in biopharmaceutical analysis.</p>

<p>Stoll is the author or co-author of more than 62 peer-reviewed publications and three book chapters in the area of separation science, is a named co-inventor on four granted patents, and has instructed numerous short courses in two-dimensional liquid chromatography. In 2011, he was the recipient of LCGC’s Emerging Leader in Chromatography Award. In 2014, he added to The Analytical Scientist’s list of ‘Top 40 Under 40’ analytical scientists.
