A team led by scientists from CEITEC Masaryk University has uncovered the infection mechanism of tailed bacteriophages – a group of viruses that infect bacteria. These phages target, among others, Staphylococcus aureus, a major cause of bacterial infections that is often resistant to antibiotics. The bacterium belongs to a group of Gram-positive bacteria characterised by an exceptionally thick cell wall. Although the infection mechanisms of some other bacteriophages have been described previously, the way tailed phages infecting Staphylococcus aureus and other thick-walled bacteria invade their hosts had remained unknown. The researchers have now revealed how such a bacteriophage breaches the protective barrier of the bacterial cell and delivers its genetic material inside. Their findings could contribute to the development of new strategies for combating antibiotic-resistant bacteria.Staphylococcus aureus is one of the most common sources of bacterial infections in humans. It can cause skin infections, pneumonia, and even life-threatening bloodstream infections. Moreover, many of its strains have become resistant to commonly used antibiotics, increasing interest in alternative treatment approaches such as bacteriophage therapy.“Bacteriophages are the most abundant biological entities on Earth and have been infecting bacteria for billions of years. Yet we still do not fully understand how they overcome the cell wall of certain Gram-positive bacteria. By reconstructing the bacteriophage structure before and after it attaches to a bacterial cell, we were able to reveal this process,” says <a target="_blank" rel="noopener noreferrer" href="https://www.ceitec.eu/jan-binovsky-ph-d/u91173?page=publication">Ján Bíňovský</a> of CEITEC Masaryk University, who led the study together with Marta Šiborová.Bacteriophage 812, investigated in this study, specifically infects Staphylococcus aureus. The researchers discovered that once it recognises the correct host, the protein structure at the tip of its tail undergoes extensive rearrangements. Individual components of this structure, which resembles a lunar landing module, reorganise into a new configuration that enables the virus to firmly attach to the bacterial surface. The bacteriophage tail contracts to approximately half of its original length, and the released energy drives the central tube through the bacterium's protective layers. The virus can then deliver its genetic material into the cell and initiate infection,” explains Marta Šiborová.Unlike many other bacteria, Staphylococcus aureus possesses an exceptionally thick cell wall. The researchers therefore investigated in detail how the bacteriophage overcomes this barrier. They found that the virus employs a combination of mechanical force and enzymatic activity. Some components of its infection apparatus likely disrupt teichoic acids, which form the outer protective layer of the bacterial cell wall. Other proteins then degrade peptidoglycan, the main structural component of the cell wall. Finally, the bacteriophage penetrates the inner lipid membrane, allowing its central tube to enter the bacterial cell.The findings show that the bacteriophage does not function as a simple "needle", but rather as a sophisticated nanomachine composed of dozens of cooperating proteins.Although this is fundamental research, the findings may prove important for future medical applications. Bacteriophages are considered one of the promising alternatives for treating infections caused by antibiotic-resistant bacteria.“If we want to use bacteriophages therapeutically in the future – or even engineer them for specific medical applications – we first need to understand in detail how they work. Rotary evaporation has long been a staple in laboratories for removing solvents from samples efficiently. However, as technology advances and the demand for higher throughput and more efficient processes increases, alternative methods are becoming more attractive. One such alternative is <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products?category=338">Organomation's S-EVAP evaporators</a>. The increasing popularity of extra virgin olive oil (EVOO) and the increasing problem of food fraud have provided the need for quality and authenticity control. Typical problems are the mislabeling of protected designation of origin (PDO) or edible oil adulteration. In this presentation we give an overview about the capabilities of different mass spectrometric methods to accurately determine the PDO of EVOO.

<li>Multi-class LC-MS and GC-MS methodologies together with statistical methods enable the discrimination between different PDOs, identifying potential origin markers.</li>
<li>The combined use of non-targeted and targeted approaches enhances the outcome of the study. GC-APCI-QTOF preserves the pseudo-molecular ion information, which is a great advantage and facilitates the identification.</li>
<li>Co-eluting isomers maybe PDO markers and these can be identified and characterized by timsTOF technology.</li>
<li>Rapid LC-free, magnetic resonance mass spectrometry (MRMS) analysis was used for mapping and quality control assessment of EVOO using statistical methods which allows for clustering according to geographical origin, harvesting year, cultivation practice and the oil production procedure.</li>

High-resolution, accurate-mass (HRAM) mass spectrometers (MS) are a class of MS instrumentation with capability to resolve complex sample matrix and to allow identification of compounds by measuring their accurate masses.
In particular, those coupled to liquid chromatograph (HRAM LC/MS) have been used extensively for structural elucidation of unknown compounds primarily in chemical industry (impurity analysis) and biological researches (proteomics, metabolomics, lipidomics), but also increasingly in forensic science, food safety and environmental testing. Given the recent reduction of instrumentational cost, it will not take many more years until HRAM analysis becomes a versatile tool for all industries and researches. 
The key performance attribute underlying all application fields is mass measurement accuracy (MMA) as it affords molecular specificity and reduction of false positive results. MMA is given in the specification of HRAM instruments in parts-per-million (ppm), frequently stating a result of a set of representative measurements e.g. “&lt;1 ppm RMS”. However, this by itself does not convey how repeatably and reproducibly this MMA can be achieved. 
The webinar describes various metrics that are relevant to statistical interpretation of MMA, which explain why mass resolution of 30,000 is more than sufficient for mass measurement at sub-ppm accuracy. Using the Shimadzu LCMS-9030 quadrupole time-of-flight MS, the rationale and strength of instrument stability for consistently achieving sub-ppm MMA has been demonstrated in small molecules.

Maximizing Mass Spectrometry Capabilities in Food Analyses for Multiresidues

There is increased concern regarding the authenticity of basmati rice throughout the world. For years, traders have been passing off a lesser quality rice as the world&#39;s finest long-grained, aromatic rice, basmati, in key markets like the US, Canada and the EU.
In this on-demand webinar, we describe a proof of principle method to assess the authenticity of basmati rice using the latest advancements in high resolution GC-MS and informatics technologies. Volatile compounds of interest were extracted from heated dry rice via SPME and headspace. Following a generic GC separation, detection was performed using a SYNAPT G2-Si MS run in HDMSE mode. Collection of a HDMSE dataset offers a high level of specificity owing to the orthogonal nature of the ion mobility separation to the GC separation.
Progenesis QI, the latest OMICS informatics package from Waters, is designed to utilize the 4-dimensional data produced during a SYNAPT G2-Si HDMSE acquisition. Initially, alignment of all injections was performed followed by the unique peak co-detection process resulting in the same number of analyte measurements in every sample and no missing values. Data from all isotopes and adducts of a parent compound were then deconvoluted giving a single robust and accurate quantitative measurement for that parent compound. The peak picking and deconvolution algorithms ensure a high quality compound table where adducts and isotopes are grouped with the parent m/z. Compounds of interest from the rice samples were highlighted using various multivariate statistical techniques such as PCA and identified using elucidation tools and relevant database searches within the software platform.
This workflow has general applicability to many areas of discovery OMICS and food research where biomarkers differentiating between two or multiple groups are required.
