At the January 2025 CFSRE Seized Drugs Symposium, a Thermo Fisher Scientific–sponsored workshop showcased the potential of <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/2240">Thermo Scientific VeriSpray PaperSpray mass spectrometry </a>(PS-MS) to transform drug analysis workflows in both forensic and clinical toxicology. Stefania’s PhD research seeks to change that. Her team is developing a rapid, extraction-free method using VeriSpray PS-MS for the targeted analysis of plasma samples from patients presenting with acute drug toxicity.By using dried matrix spots on a paper substrate and coupling this with a <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/404">Thermo Scientific TSQ Altis Plus Triple Quadrupole Mass Spectrometer</a>, Stefania’s work provides an alternative screening technique to chromatography, delivering faster, actionable results to guide early clinical decision-making.<h5>Key takeaways:</h5><ul><li>Paper spray MS is an ambient ionization technique suitable for emergency toxicology due to its rapid results and minimal sample prep.</li><li>Enhancing the paper substrate and adding microextraction steps can boost sensitivity without sacrificing speed.</li></ul><h3>Substrate matters: boosting detection with paper treatment</h3>One key limitation of PS-MS is the interference that can arise from the paper substrate itself. Marwa’s research explored nine different paper treatments, ranging from acids to solvents, to improve the signal-to-blank (S/B) ratio when analyzing eight common drugs in whole blood.The most promising results came from formic acid and nitric acid treatments, which significantly enhanced analyte detection. Although these methods are currently time-consuming, Marwa envisions future industrial-scale production of pre-treated cartridges to support scalable clinical workflows.<h5>Key takeaways:</h5><ul><li>Substrate optimization can significantly reduce background noise in PS-MS analysis.</li><li>Acid-treated paper enhances S/B ratios, enabling more reliable detection of drugs in blood.</li><li>Industrial pre-treatment could make these improvements broadly accessible.</li></ul><h3>DLLME meets paper spray: improving sensitivity in plasma</h3>Saeed Almheiri’s presentation focused on dispersive liquid-liquid microextraction (DLLME) as a sample prep technique to increase sensitivity for PS-MS analysis of drugs in plasma, without adding significant complexity to the workflow.Using DLLME prior to VeriSpray analysis, Saeed demonstrated improved detection limits (as low as 0.3 ng/mL) across a panel of 10 drugs, including buprenorphine, cocaine, and MDMA. Importantly, this method maintained the fast, minimal-prep advantages that make paper spray ionization so appealing. She drives the implementation of ICH Q2(R2), Q14, and Analytical Quality by Design (AQbD) principles in collaboration with global biopharmaceutical partners. Amanda serves as a member of the United States Pharmacopeia (USP) Pharmaceutical Analytical Lifecycle and Data Science Expert Committee and is a board member of the ECA Analytical Quality Control Group. Before joining RIC, she spent more than 12 years at USP, where she worked both as a Senior Scientist – leading method development and validation projects, impurity analysis by LC-MS/GC-MS, and reference standards characterization by LC-HRMS – and as a Scientific Affairs Manager, contributing to major standards such as &lt;1220&gt; and &lt;1225&gt; and driving initiatives in AQbD, predictive modeling, and analytical procedure lifecycle management. Drug testing can be performed in many biological matrices, such as urine, blood, saliva, sweat, hair and nail. Although urine and blood testing are the most common forms of drug testing, the use of keratinized matrices such as hair and nail is becoming extremely valuable in providing a broader picture of past drug consumption.
This is due to the incorporation of drugs consumed by the subject in the hair and nail keratin matrices, offering a much wider detection window than conventional biological matrices. In addition to the non-invasive nature of sample collection, hair and nail specimens are easy to transport and store, which minimizes the risk of sample alteration and degradation overtime. Despite its rise in popularity, drug detection in keratinized matrices remains challenging due to the low concentration of drugs embedded in the keratin matrix. As a result, hair and nail analysis requires sensitive instrumentation and extensive validation. 
This webinar will focus on the method development and validation of LC-MS/MS-based methods required to perform comprehensive and sensitive drug analysis in keratinized matrices. Case examples of the use of these methods will be demonstrated, highlighting the need for high-sensitivity mass spectrometry instrumentation for low-level detection of drugs and metabolites in hair and nail samples. The result presented will highlight the benefits of hair and nail analyses in forensic applications, enabling researchers to obtain significant retrospective information about past exposure and long-term use of drugs in the consumer’s population.

Mass spectrometry is a commonly employed platform for quantifying analytes. For polymers and complicated mixtures, the lack of standards and the difficulty in representing the large intact molecule make quantitation difficult if not impossible.In this presentation, the quantitation of previously unquantifiable analytes will be described. These analytes include polysaccharides, large protein mixtures and their posttranslational modifications, and complicated mixtures such as oligosaccharides in human milk.Polysaccharides are digested by a chemical enzyme and the resulting oligosaccharides are used to quantitate the intact polysaccharide using LC-MS and dynamic MRM methods. Human milk oligosaccharides are quantified using a selected number of standards and interpolating the abundances of those without standards to obtain the absolute abundances of nearly a hundred analytes.Protein mixtures are quantified using total protein quantification method while further employing these values with selected protein standards to obtain absolute quantitation of hundreds of proteins in bovine milk. Additionally, quantitation of glycan modification on proteins using dynamic MRM monitors the glycosylation of individual sites on proteins in serum.<blockquote>Presenter: &nbsp;Carlito Lebrilla, Ph.D. (Professor of Analytical Chemistry, University of California at Davis)Dr. Carlito B. He was an Alexander von Humboldt Fellow and a NSF-NATO Fellow at the Technical University in Berlin. He returned to the UC Irvine as a President’s Fellow and has been at UC Davis since 1989. He has served as Chair of the Chemistry Department. His research is in Analytical Chemistry, primarily mass spectrometry with applications to clinical glycomics and biofunctional food. He has nearly 400 peer-reviewed publications. He is also Co-Chief editor of Mass Spectrometry Reviews and has been on the editorial board of Molecular and Cellular Proteomics, Mass Spectrometry Reviews, Journal of American Society for Mass Spectrometry, European Mass Spectrometry, and International Journal of Mass Spectrometry.</blockquote>
